摘要
目的探讨环氧化酶(COX)-2、增殖细胞核抗原Ki-67在前列腺癌组织中的表达水平及其临床意义。方法选择该院2017年1月至2019年6月收治的前列腺癌患者67例为观察组,选取同期良性前列腺疾病患者81例为对照组。分别采用免疫组织化学法、荧光定量PCR检测COX-2和Ki-67水平及mRNA相对表达量;采用蛋白质印迹法分析2组前列腺组织标本中COX-2和Ki-67蛋白表达灰度值差异。结果观察组COX-2阳性率为71.64%,Ki-67阳性率为67.16%,对照组分别为16.05%、11.11%,差异有统计学意义(χ^2=46.774,P<0.05;χ^2=49.716,P<0.05)。观察组COX-2 mRNA相对表达量为1.68±0.51,Ki-67 mRNA相对表达量为0.83±0.15,对照组依次为0.32±0.10、0.12±0.03,差异有统计学意义(t=23.476,P<0.05;t=41.633,P<0.05)。观察组COX-2蛋白表达灰度值为0.76±0.15,Ki-67蛋白表达灰度值为0.58±0.14,均显著高于对照组的0.27±0.05、0.16±0.03,差异有统计学意义(t=27.620,P<0.05;t=25.145,P<0.05)。COX-2、Ki-67表达水平与前列腺癌患者临床T分期、淋巴结转移及血清前列腺特异抗原(PSA)水平关系密切(P<0.05)。结论COX-2和Ki-67过表达与前列腺癌的发生有关,二者可成为前列腺癌的诊断标志物和潜在治疗靶点。
Objective To investigate the expression and clinical significance of cyclooxygenase(COX)-2 and proliferating cell nuclear antigen Ki-67 in prostate cancer.Methods Sixty-seven patients with prostate cancer admitted to the hospital from January 2017 to June 2019 were selected as the observation group,and 81 patients with benign prostate disease in the same period were selected as the control group.COX-2 and Ki-67 levels and mRNA relative expression were detected by immunohistochemistry and fluorescence quantitative PCR respectively,and the gray value difference of COX-2 and Ki-67 protein expression was detected by Western blot.Results The positive rate of COX-2 in the observation group was 71.64%,the positive rate of Ki-67 was 67.16%,and the control group was 16.05%,11.11%,the difference was statistically significant(χ^2=46.774,P<0.05;χ^2=49.716,P<0.05).COX-2 mRNA relative expression was 1.68±0.51 in the observation group,Ki-67 mRNA relative expression was 0.83±0.15 in the control group,and 0.32±0.10 and 0.12±0.03 in the control group,the difference was statistically significant(t=23.476,P<0.05;t=41.633,P<0.05).The gray values of COX-2 protein and Ki-67 protein in the observation group were 0.76±0.15 and 0.58±0.14,which were significantly higher than those in the control group(0.27±0.05,0.16±0.03),the difference was statistically significant(t=27.620,P<0.05;t=25.145,P<0.05).The expression of COX-2 and Ki-67 was closely related to clinical T stage,lymph node metastasis and serum PSA level in patients with prostate cancer(P<0.05).Conclusion The overexpression of COX-2 and Ki-67 is related to the occurrence of prostate cancer,and they can be the diagnostic markers and potential therapeutic targets of prostate cancer.
