摘要
为了进一步挖掘黄姑鱼的基因数据库资源,本研究采用高通量测序技术对采自舟山的黄姑鱼进行了全转录组测序,获取了42 809 266条Clean Reads。Clean Reads通过从头组装并进一步聚类为32 623条Unigenes,平均长度和N50分别为1 646 bp和2 777 bp。利用国际上7个主要的基因注释数据库(Nr、Nt、Pfam、KOG、Swiss-prot、KEGG和GO)对所有的Unigenes进行基因功能分析,共有28 645条Unigenes在至少以上一个数据库中成功获得注释信息。而后将所有的Unigenes进行蛋白库比对和estscan软件预测,共获得30 382个CDS;分子标记筛查后共得到了29 022个潜在的SSRs。作为一项重要的研究基础,本研究提供的转录组信息有助于为黄姑鱼的分子水平研究提供新的认识。
In order to enrich the basic genetic database resource of Nibea albiflora, we obtained a transcriptome data of N. albiflora from combined tissues(liver, ovary, gill, eye and muscle) using Illumina RNA-seq technology. A total of 62 879 204 clean reads were obtained from sequencing platform. All clean reads were de novo assembly and further clustered to produce 32 623 unigenes, the mean length and N50 of all unigenes was1 646 bp and 2 777 bp, respectively. Among all the unigenes, a total of 28 645 unigenes were obtained annotation information in at least one protein database(Nr, Nt, Pfam, KOG, Swiss-prot, KEGG and GO). Furthermore, 30 382 CDS were predicted by blast with protein databases and estscan software. Putative molecular markers were identified in our dataset and 24 443 SSRs were obtained in the present study. As an important research foundation, this study can provide a novel understanding for the molecular aspects of N. albiflora.
