检测血清标志物联合定量MRI评估胫骨平台软骨体积在骨性关节炎犬模型中的诊断价值 被引量:4

Diagnostic value of combined serum markers and quantitative MRI evaluation of cartilage volume of tibial plateau in a surgically-induced osteoarthritis dog model

摘要 目的评估血清Ⅱ型胶原羧基端端肽(CTX-Ⅱ)和X型胶原蛋白(COL10A1)水平联合定量MRI评估胫骨平台软骨体积在骨性关节炎犬模型中的诊断价值。方法选择18只健康雄性比格犬,随机分为2组,10只通过右膝前交叉韧带横行离断法建立骨性关节炎模型(模型组),8只进行假手术(对照组)。比较模型组与对照组术后4、8、12、16周CTX-Ⅱ、COL10A1水平,以及术前、术后4周、术后8周、术后16周MRI图像上右侧胫骨平台软骨体积。结果模型组与对照组术后4周血清CTX-Ⅱ水平差异无统计学意义(P>0.05);模型组术后8、12、16周血清CTX-Ⅱ水平高于对照组,差异有统计学意义(P<0.05)。模型组与对照组术后4周、术后16周血清COL10A1水平差异无统计学意义(P>0.05);模型组术后8周、12周血清COL10A1水平高于对照组,差异有统计学意义(P<0.05)。模型组与对照组术前、术后4周胫骨平台软骨体积差异无统计学意义(P>0.05);模型组术后8周、16周胫骨平台软骨体积小于对照组,差异有统计学意义(P<0.05)。结论检测血清CTX-Ⅱ和COL10A1变化联合定量MRI评估胫骨平台软骨体积在骨性关节炎犬模型中具有反映和监测骨性关节炎进展的潜力。 Objective To assess the diagnostic value of serum C-terminal telopeptide of typeⅡcollagen(CTX-Ⅱ)and Collagen X combined with quantitative MRI evaluation of cartilage volume of tibial plateau in a canine model of osteoarthritis.Methods Eighteen male Beagle dogs were randomly divided into two groups.Osteoarthritis models(model group)were surgically induced via anterior cruciate ligament transection(ACLT)of the right knee in ten beagles,and eight beagles were used for sham operation(control group).The levels of CTX-Ⅱ,COL10 A1 at 4,8,12,and 16 weeks after operation were compared between the model group and the control group.The volume of cartilage on the right tibial plateau on the MRI images before operation and 4,8,16 weeks after operation was compared between two group.Results There was no significant difference in serum CTX-Ⅱlevels between the model group and the control group at 4 weeks after operation(P>0.05).The levels of serum CTX-Ⅱin the model group were higher than those in the control group at 8,12,and 16 weeks after operation(P<0.05).There was no significant difference in serum COL10A1 levels between the model group and the control group at 4 weeks and 16 weeks after surgery(P>0.05).The levels of serum COL10A1 in the model group at 8 and 12 weeks after operation were higher than those in the control group,with a significant difference(P<0.05).There was no significant difference in the tibial platelet cartilage volume between the model group and the control group before and 4 weeks after operation(P>0.05).The tibial platelet cartilage volume in the model group at 8 and 16 weeks after surgery was smaller than that in the control group,and the difference was statistically significant(P<0.05).Conclusion Detection of changes in serum CTX-Ⅱand COL10A1 combined with quantitative MRI evaluation of cartilage volume of tibial plateau in dog models of osteoarthritis have the potential to reflect and monitor the progression of osteoarthritis progression.
