摘要
目的建立无髓犬年轻恒牙牙髓血运重建动物模型,观察富血小板血浆(platelet-rich plasma,PRP)对年轻恒牙牙髓血运重建的作用。方法选取6例5月龄健康杂种犬,将上、下颌第一、二、三前牙(牙根未发育完成)拔除牙髓组织。观察组在根管内加入PRP进行牙髓血运重建,对照组不加PRP进行牙髓血运重建。3个月后处死各动物,组织切片HE染色观察新生组织形态,蛋白免疫印记(Western blot)检测成牙本质/成骨细胞标志物牙本质涎蛋白(dentin sialoprotein,DSP)表达情况。结果根管内均有新生组织形成,组织学观察主要为纤维样组织、骨样组织及牙骨质样组织;观察组DSP表达高于对照组。结论牙髓血运重建中新生组织主要为纤维样组织、骨样组织及牙骨质样组织;PRP对牙髓血运重建中细胞增殖及分化具有促进作用。
Objective To establish dog model of pulp revascularization in pulpless immature permanent teeth and to observe the effect of platelet-rich plasma on pulp revascularization.Methods Six 5-month old dogs were used.In the observation group,platelet-rich plasma(PRP) was added to the root canal for pulp revascularization,while in the control group,no PRP was added to the root canal for pulp revascularization.3 months later,the animals were sacrificed.HE staining of tissue sections was used to observe the morphology of new tissue and Western blot was used to detecte the expression of DSP.Results Both groups had new tissue formation in the root canal.The regenerated tissues in the root canals after revascularization were mainly fiber-like,bone-like and cementum-like tissue.The DSP expression was higher in the observation group than in the control group.Conclusion The regenerated tissues in the root canals after revascularization were mainly fiber-like,bone-like and cementum-like tissue.PRP had obvious effects on proliferation and differentiation of cells in pulp revascularization.
