丹红注射液静注对大鼠肾病的治疗作用及其可能机制 被引量:3

摘要 目的观察丹红注射液对大鼠肾病的治疗作用,并探讨其可能机制。方法32只健康雄性SD大鼠随机分为4组各8只,丹红组、氯沙坦组、模型组均静注6 mg/kg阿霉素制备肾病模型,对照组不制备模型;制模第4周,丹红组静注4 mL/(kg·d)的丹红注射液,氯沙坦组灌胃给予12 mg/(kg·d)的氯沙坦,模型组和对照组灌胃给予12 mg/(kg·d)的生理盐水,连续4周。比较各组尿液尿蛋白及血清白蛋白(Alb)、TG、TC、尿素氮、肌酐、生长分化因子(GDF-15)和肾组织肾小球硬化指数、GDF-15蛋白、GDF-15 mRNA。结果与对照组比较,其余3组尿蛋白、TG、TC、尿素氮、肌酐、肾小球硬化指数升高,Alb降低(P均<0.05);与模型组比较,丹红组和氯沙坦组尿蛋白、TG、TC、尿素氮、肌酐、肾小球硬化指数降低,Alb升高(P均<0.05);与氯沙坦组比较,丹红组尿蛋白、尿素氮、肌酐、肾小球硬化指数降低,Alb升高(P均<0.05)。与对照组比较,其余3组血清GDF-15水平及肾组织GDF-15蛋白、mRNA表达升高(P均<0.05);与模型组比较,丹红组和氯沙坦组血清GDF-15水平及GDF-15蛋白、mRNA表达降低(P均<0.05);与氯沙坦组比较,丹红组血清GDF-15水平及GDF-15蛋白、mRNA表达降低(P均<0.05)。结论丹红注射液静注对大鼠肾病有治疗作用,其机制可能通过抑制血清GDF-15及肾组织GDF-15 mRNA、蛋白表达,进而减少尿蛋白,最终减轻大鼠肾脏病理损害。 Objective To observe the therapeutic effect of Danhong injection on nephropathy in rats and to explore its possible mechanism.Methods Thirty-two healthy male SD rats were randomly divided into 4 groups:Danhong group,losartan group,model group,and control group,with 8 rats in each.Rats in the Danhong group,losartan group,and model group were intravenously injected with 6 mg/kg adriamycin to prepare the nephropathy models,while rats in the control group were not treated.At the 4th week,rats in the Danhong group were intravenously injected with 4 mL/(kg·d)Danhong injection,losartan group with 12 mg/(kg·d)losartan,model group and control group with 12 mg/(kg·d)saline for 4 weeks.Urine protein,serum albumin(Alb),triglyceride(TG),total cholesterol(TC),urea nitrogen,creatinine,growth differentiation factor(GDF-15),glomerulosclerosis index,GDF-15 protein and mRNA were compared between groups.Results Compared with the control group,the urine protein,TG,TC,urea nitrogen,creatinine,and glomerulosclerosis index increased and Alb decreased in the remaining three groups(all P<0.05).Compared with the model group,the urine protein,TG,TC,urea nitrogen,creatinine,and glomerulosclerosis index decreased and Alb increased in the Danhong group and losartan group(all P<0.05);compared with the losartan group,the urine protein,urea nitrogen,creatinine,and glomerulosclerosis index of the Danhong group decreased,and Alb increased(all P<0.05).Compared with the control group,the level of serum GDF-15 and the expression levels of GDF-15 protein and mRNA in the renal tissues increased in the other three groups(all P<0.05).Compared with the model group,the serum level of GDF-15 and the expression levels of GDF-15 protein and mRNA decreased in the Danhong group and losartan group(all P<0.05).Compared with the losartan group,the serum GDF-15 level and the expression levels of GDF-15 protein and mRNA in the Danhong group decreased(all P<0.05).Conclusion The intravenous injection of Danhong injection has a therapeutic effect on nephropathy rats through inhibiting the expression of serum GDF-15 and GDF-15 mRNA and protein in the renal tissues,and thus reducing urine protein and alleviating renal pathological damage.
