基于生物信息学分析P-Rex1在胃癌组织中的表达及临床意义 被引量:1

摘要 目的:分析P-Rex1在胃癌肿瘤组织中的表达,探讨其在胃癌发展过程中的临床意义及其可能的调控网络。方法:利用TCGA胃癌数据库分析P-Rex1在胃癌肿瘤组织和癌旁组织中的表达差异,并分析其在不同肿瘤进展阶段的表达差异以及其与幽门螺旋杆菌感染的相关性;利用Oncomine和Kaplan-Meier Plotter工具分析P-Rex1表达与胃癌患者预后的关系,并阐述其与胃癌临床病理学参数的相关性;基于cBioPortal工具分析TCGA胃癌患者肿瘤组织P-Rex1基因的共表达网络,进行GO聚类分析以及蛋白-蛋白相互作用网络分析。结果:胃癌肿瘤组织中的P-Rex1基因表达显著增加;P-Rex1基因表达水平与肿瘤分期存在正相关的联系,但与幽门螺旋杆菌感染与否无显著相关性;高表达P-Rex1的胃癌患者总生存期和进展后生存期都显著下降;HER2阴性胃癌患者中P-Rex1高表达组总生存期显著低于P-Rex1低表达组患者;伴有淋巴结转移的胃癌患者P-Rex1高表达组总生存期显著低于P-Rex1低表达组;P-Rex1蛋白质相互作用共表达网络发现P-Rex1与CDC42、Rac分子和系列Rho信号分子存在紧密联系,通过聚类分析发现P-Rex1可能参与淋巴细胞的激活、细胞因子释放、炎症免疫信号通路等过程。结论:P-Rex1在胃癌组织中高表达,其表达水平与胃癌发展过程存在紧密联系,生存期分析发现在HER2阴性或淋巴结转移胃癌患者中,P-Rex1表达与患者预后存在紧密的联系,并且可能参与胃癌的炎症免疫信号网络调节。 Objective:To analyze the expression levels of P-Rex1 in gastric cancer,explore its clinical significance during the cancer development and its potential regulation network.Methods:Using TCGA gastric cancer data to test the expression difference in gastric tumor tissues and surrounding tissues,exploring the correlation with tumor stage,tumor grade and H.pylori infection.With the Oncomine and Kaplan-Meier Plotter tool to analyze the association between P-Rex1 expression and prognosis.Based on the application of cBioPortal to analyze the co-expression network of P-Rex1 in TCGA gastric cancer,GO cluster analysis and protein-protein interaction were also carried based on the co-expression network.Results:P-Rex1 was high increased in gastric tumor tissues,and P-Rex1 expression was positive correlated with the tumor stage and tumor grade.Besides,the expression of P-Rex1 had no significant correlation with H.pylori infection.The high expression in gastric cancer predicted the poor prognosis including overall and post progression survival.In the HER2 negative or lymph node metastasis positive gastric cancer patients,the high expression of P-Rex1 showed significant lower survival rate.The protein-protein interaction and co-expression network of P-Rex1 showed P-Rex1 had closely associated with CDC42,Rac and Rho signaling network,and GO analysis showed P-Rex1 had potential role in the process of lymphocyte activation,cytokine production and immune inflammation signaling pathway.Conclusion:P-Rex1 is highly expressed in gastric cancer tissues,its expression level has closely associated with development of gastric cancer.Survival rate analysis showed the P-Rex1 expression has closely associated with prognosis in HER2 positive and lymph node metastasis,and possibly participate in the immune inflammation regulation of gastric cancer.
