壮骨止痛方对去势雌鼠转化生长因子β1 mRNA及Ⅰ型胶原纤维mRNA表达的影响

Effect of Zhuanggu Zhitong Prescription(壮骨止痛方)on Expression of Transformed Growth Factorβ1 mRNA and TypeⅠCollagen Fiber mRNA in Castrated Female Rats

目的:观察壮骨止痛方对去势雌鼠转化生长因子β1(Transforming growth factor-β,TGF-β1)mRNA及Ⅰ型胶原纤维mRNA表达的影响,并探究其治疗骨质疏松的具体机制。方法:将75只健康雌性SD大鼠随机分为空白组、假手术组、模型组、壮骨止痛方组、无水乙醇提取A部位组,每组15只。除空白组、假手术组外,其余各组建立绝经后骨质疏松病理模型,假手术组去除卵巢周围相应体积脂肪。造模后5 d开始给药,13周后取股骨,原位杂交法检测分析骨组织中Ⅰ型胶原纤维mRNA和TGF-β1 mRNA的表达。结果:模型组TGF-β1 mRNA和Ⅰ型胶原mRNA表达明显低于空白组(P<0.05),壮骨止痛方组、无水乙醇提取A部位组TGF-β1 mRNA和Ⅰ型胶原纤维mRNA表达明显高于模型组,差异有统计学意义(P<0.05)。结论:壮骨止痛方可能是通过提高TGF-β1 mRNA和Ⅰ型胶原纤维mRNA的表达,促进成骨细胞分化,发挥改善骨质疏松的效应。

Objective:To observe the effect of alcohol extract A of Zhuanggu Zhitong Prescription(ZGZFP)on the expression of TGF-β1 mRNA and typeⅠcollagen fiber mRNA in defoliated female mice,and to explore the specific mechanism of its treatment of osteoporosis.Methods:The 75 healthy female SD rats were randomly divided into blank group,sham operation group,model group,ZGZTP group and anhydrous ethanol extraction group,with 15 rats in each group.In addition to the blank group and the sham group,pathological models of postmenopausal osteoporosis were established in the remaining groups,and corresponding volume fat around the ovary was removed in the sham group.The drug was administered 5 days after the operation,and the femur was taken at 13 weeks to analyze the expression of typeⅠcollagen mRNA and TGF-β1 mRNA in bone tissue.Results:The expressions of TGF-β1 mRNA and typeⅠcollagen mRNA in the model group were significantly decreased by in situ hybridization,and there were significant differences between the control group and the blank group(P<0.05).The expression levels of the whole formula group and the alcohol extract A group were significantly increased compared with the model group,and the difference was statistically significant(P<0.05).Conclusion:Zhuanggu Zhitong prescription may promote the differentiation of osteoblasts and improve the effect of osteoporosis by increasing the expression of TGF-β1 mRNA and typeⅠcollagen fiber mRNA.
