白细胞介素25、白细胞介素33和胸腺基质淋巴细胞生成素在慢性鼻-鼻窦炎伴鼻息肉中的表达及意义 被引量:2

Expression and clinical implications of IL-25,IL-33 and TSLP in eosinophilic and non-eosinophilic chronic rhinosinusitis with nasal polyps

摘要 目的分析白细胞介素25(interleukin-25,IL-25)、白细胞介素33(interleukin-33,IL-33)和胸腺基质淋巴细胞生成素(thymic stromal lymphopoietin,TSLP)在慢性鼻-鼻窦炎伴鼻息肉(chronic rhinosinusitis with nasal polyps,CRSwNP)患者中的表达及意义。方法应用免疫组织化学技术和实时荧光定量PCR技术分别在蛋白和mRNA水平检测IL-25、IL-33和TSLP在33例嗜酸细胞性慢性鼻-鼻窦炎伴鼻息肉(eosinophilic CRSwNP,ECRSwNP)、30例非嗜酸细胞性慢性鼻-鼻窦炎伴鼻息肉(non-ECRSwNP)和20例正常下鼻甲组织(对照组)中表达。结果IL-25mRNA和蛋白在ECRSwNP组和non-ECRSwNP组表达均高于对照组(P<0.05);IL-33 mRNA和蛋白在ECRSwNP组、non-ECRSwNP组和对照组表达差异无统计学意义(P>0.05);TSLP mRNA和蛋白在ECRSwNP组表达高于non-ECRSwNP组和对照组(P<0.05)。结论IL-25、IL-33和TSLP在ECRSwNP及non-ECRSwNP均有表达;IL-25和TSLP在CRSwNP患者鼻息肉中表达显著增高,在CRSwNP患者鼻息肉发病机制中发挥重要的作用。 OBJECTIVE Toevaluate the expression and clinical implications of IL-25,IL-33 and TSLP in patients with eosinophilic chronic rhinosinusitis with nasal polyps(ECRSwNP)and non-eosinophilic chronic rhinosinusitis with nasal polyps(non-ECRSwNP).METHODS Immunohistochernistry and quantitative RT-PCR were used to detect the mRNA and protein expression of IL-25,IL-33 and TSLP in 33 subjects with ECRSwNP,30 subjects with non ECRSwNP and 20 normal inferior turbinate tissues(control group).RESULTS The mRNA and protein expression level of IL-25 was significantly higher in the ECRSwNP and non-ECRSwNP groups than that in the control group(P<0.05,P<0.01);The mRNA and protein expression level of IL-33 was not significantly different among the three groups(F>0.05);The TSLP mRNA and protein expression was significantly higher in the ECRSwNP group than that in the non-ECRSwNP and control groups(P<0.05,P<0.01).CONCLUSION The IL-25,IL-33 and TSLP were both expressed in eosinophilic and non-eosinophilic chronic rhinosinusitis with nasal polyps,meanwhile the expression of IL-25 and TSLP elevate in CRSwNP.The IL-25 and TSLP plays an im portant role in the pathogenesis of CRSwNP.
