摘要
目的评价担载万古霉素的水凝胶缓释药剂对耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcus aureus,MRSA)的体外抑菌效果。方法制备担载万古霉素的水凝胶,确定其对MRSA的最低抑菌浓度(minimum inhibitory concentration,MIC)和最低杀菌浓度(minimum bactericidal concentration,MBC),探讨其对MRSA的抑菌杀伤效应及生物菌膜生物量和细胞活性的影响。结果与未成胶的万古霉素相比,万古霉素水凝胶对MRSA的MIC(1μg/mL)和MBC(2μg/mL)分别各降低了1倍。对于MRSA的抑菌杀伤效应,万古霉素水凝胶呈缓慢释放并持续抑菌。万古霉素水凝胶对MRSA成熟生物菌膜生物量无显著影响,能增强对生物菌膜细胞活性的杀伤并减少万古霉素使用量,具有很好的抑菌杀菌效果。结论担载万古霉素水凝胶在减少用药剂量下与万古霉素抑菌效果相当,本研究为后续研发生物利用度好的新型缓释万古霉素药剂提供了参考。
Objective To evaluate the in vitro antibacterial effect of sustained release hydrogel loaded with vancomycin on methicillin-resistant Staphylococcus aureus(MRSA). Methods The hydrogel loaded with vancomycin was prepared,of which the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)to MRSA were determined,and the influences on the bacteriostatic and killing effects,biofilm biomass and cell activity of MRSA were investigated.Results Compared with those of vancomycin,either the MIC(1 μg/mL)or the MBC(2 μg/mL)of hydrogel loaded with vancomycin to MRSA decreased by one fold. The hydrogel loaded with vancomycin was slowly released, which continuously inhibited Staphylococcus aureus. However,the hydrogel showed no significant influence on the mature biofilm biomass of MRSA,while enhanced the killing activity of biofilm cells and reduced the dose of vancomycin,which showed good antibacterial and killing effects. Conclusion The bacteriostatic effect of hydrogel loaded with vancomycin was comparable to that of vancomycin at decreased dosage,which provided a reference for the subsequent development of novel sustained release form of vancomycin.
