乳酸菌制剂对环磷酰胺致大鼠免疫抑制的调节作用

摘要 目的观察嗜酸乳杆菌SD65、干酪乳杆菌SD07和植物乳杆菌复合制剂对环磷酰胺所致免疫损伤大鼠的免疫功能调节作用。方法采用环磷酰胺(CTX)致免疫低下大鼠模型,灌胃乳酸菌制剂,观察其对大鼠体重、胸腺指数、脾脏指数、部分固有免疫细胞、CD4/CD8及血清IL-6、TNF-α含量和脾脏IL-2、IL-6、TNF-α基因表达水平的影响。结果给药第7天~药后27天,乳酸菌制剂1.25,2.5,5.0 ml/kg干预组动物体重高于模型组(P<0.01或P<0.05);给药结束时乳酸菌制剂干预组胸腺指数、脾脏指数高于模型组(P<0.05);乳酸菌制剂5.0 ml/kg干预组白细胞数目高于模型组(P<0.05),1.25,2.5,5.0 ml/kg干预组CD4/CD8均高于模型组(P<0.05或P<0.01)。乳酸菌制剂2.5,5.0 ml/kg干预组血清IL-6含量高于模型组(P<0.05),1.25,2.5,5.0 ml/kg干预组血清TNF-α含量低于模型组(P<0.05或P<0.01)。乳酸菌制剂2.5,5.0 ml/kg干预组IL-2基因表达水平高于模型组(P<0.05),2.5 ml/kg干预组IL-6基因表达水平高于模型组(P<0.05),作用呈剂量依赖性,高剂量组的TNF-α基因表达水平显著低于模型组(P<0.05)。结论乳酸菌制剂可明显提高CTX损伤所致免疫低下大鼠的免疫功能。 Objective To observe the regulatory effect of Lactobacillus acidophilus SD65,Lactobacillus casei SD07 and Lactobacillus plantarum oral compound preparation on immunosuppression in rats induced by cyclophosphamide(CTX).Methods Hypoimmunity in rats were induced by intraperitoneal injection of CTX.The Lactobacillus preparation was given to rats by gavage,and its effects on body weights,thymus index,spleen index,some innate immune cells,CD4/CD8,serum levels of IL-6 and TNF-α,as well as gene expression levels of IL-2,IL-6 and TNF-α in spleen were observed.Results From day 7 to day 27 after administration,the body weights of the animals treated with Lactobacillus preparation(1.25,2.5,5.0 ml/kg)were higher than those of the animals in the model group(P<0.01 or P<0.05).At the end of administration period,the thymus index and spleen index of the animals treated with Lactobacillus preparation were higher than those of the animals in the model group(P<0.05).The number of white blood cells of the animals treated with Lactobacillus preparation(5.0 ml/kg)was higher than that of the animals in the model group,and CD4/CD8 of Lactobacillus preparation(1.25,2.5,5.0 ml/kg)treated animals was also higher(P<0.01 or P<0.05).The serum level of IL-6 of the Lactobacillus bacteria preparation(2.5,5.0 ml/kg)treated animals was higher than that of the model group(P<0.05),and the serum level of TNF-αof the Lactobacillus bacteria preparation(1.25,2.5,5.0 ml/kg)treated animals was lower than that of the model group(P<0.05 or P<0.01).The Lactobacillus bacteria preparation(2.5,5.0 ml/kg)treated animals showed significantly higher gene expression levels of IL-2 than those of the animals in the model group(P<0.05),and Lactobacillus bacteria preparation(2.5 ml/kg)treated animals showed significantly higher gene expression level of IL-6 than that of the model group(P<0.05).The effects were dose-dependent.TNF-αgene expression level of Lactobacillus bacteria preparation(5.0 ml/kg)group was significantly lower than that of the model group(P<0.05).Conclusion Lactobacillus preparation can significantly improve the immune function of immunocompromised rats induced by CTX.
