摘要
目的利用紫外(UV)和常压室温等离子体(ARTP)复合诱变技术筛选甲萘醌-7(MK-7)高产菌株。方法以产MK-7的纳豆芽孢杆菌为出发菌株,利用UV+ARTP复合诱变筛选MK-7高产菌株,并对突变菌株的遗传稳定性进行研究。结果从初筛的77株突变株中获得一株MK-7高产菌株UA31#,其产量由出发菌株的7.8 mg/L提高到20.5 mg/L,经12代传代培养性能稳定。结论通过UV+ARTP复合诱变可获得遗传稳定性良好的高产MK-7菌株。
Objective To screen a high-yield MK-7-producing strain mutated by ultraviolet(UV)combined with atmospheric and room temperature plasma(ARTP).Methods Using MK-7-producing Bacillus natto as the starting strain,UV-ARTP compound mutagenesis was adopted to screen high-yield MK-7-producing strain,and the genetic stability of the mutant strain was also studied.Results A high-yield MK-7 strain UA31# was obtained from 77 mutants screened preliminarily.The yield of UA31# increased from 7.8 mg/L of the starting strain to 20.5 mg/L,and its performance was stable after 12 generations of subculture.Conclusion The high-yield MK-7-producing strain with good genetic stability can be obtained via UV-ARTP compound mutagenesis.
