摘要
为了解药用植物川续断中热激蛋白基因的分布情况及序列特征,以不同海拔川续断转录组数据库为基础,利用生物信息学方法鉴定和分析川续断HSP70和HSP90基因家族成员。结果表明,川续断中至少包含36个HSP70和18个HSP90基因,各成员在细胞质、细胞核、内质网中均有分布,15类保守基序长度介于15~50个氨基酸之间;系统进化树揭示DaHSP90基因与拟南芥亲缘关系更近。初步筛选了10个响应温度胁迫的热激蛋白基因,为进一步探索川续断HSP基因家族的功能奠定重要理论基础,同时也为研究影响其活性成分川续断皂苷Ⅵ含量因素提供参考。
In order to understand the distribution and sequence characteristics of heat shock protein genes in D.asper,which was a medicinal plant,bioinformatics methods were used to identify and analyze the members of the DaHSP70 and DaHSP90 gene families based on transcriptome databases of D.asper at different altitudes.The results showed that a total of 36 HSP70 and 18 HSP90 genes were identified from D.asper.The members were distributed in cytoplasm,nucleus or endoplasmic reticulum.The length of 15 conserved motifs of family members ranged from 15 to 50 amino acids.Phylogenetic tree revealed that DaHSP90 was closer to Arabidopsis thaliana.In this study,ten heat shock protein genes were preliminarily screened.The results provided an important theoretical foundation to explore the function of HSP gene family and study the factors affecting the content of active ingredient asperosaponinⅥin D.asper.
