葛根素通过调控IL-6相关通路诱导宫颈癌细胞凋亡

Puerarin Induces Apoptosis of Cervical Cancer Cells by Regulating IL-6 Related Pathway

目的探讨葛根素对宫颈癌Hela细胞恶性行为学的抑制作用及相关机制。方法通过CCK-8法测定0、10、20、40μmol/L浓度的葛根素作用于宫颈癌Hela细胞后增殖能力的变化,通过流式细胞术检测细胞凋亡率,Elisa检测细胞培养上清液中炎症相关因子IL-6表达情况,Western blot检测相关蛋白表达情况。结果与对照组比较,葛根素处理组Hela细胞增殖能力被显著抑制,细胞凋亡率明显升高,细胞培养环境中炎症相关因子IL-6的表达显著下调。结论葛根素对宫颈癌增殖等恶性行为学具有显著抑制作用,可能通过调控炎症因子IL-6活性起作用。

Objective To investigate the inhibitory effect of puerarin on Hela cells and its mechanism. Methods The proliferation of Hela cells was determined by CCK-8 method with concentrations of 0,10,20 and 40 mol/L puerarin.Apoptosis rate was detected by flow cytometry,the expression of inflammatory factor IL-6 in supernatant was detected by ELISA,and the expression of related proteins was detected by western blot.Results Compared with the control group,the proliferation ability of Hela cells in the puerarin treatment groups was significantly inhibited,the apoptosis rate was significantly increased,and the expression of inflammation-related factor IL-6 in supernatant was significantly down-regulated.Conclusion Puerarin has a significant effect on the proliferation of cervical cancer cells by regulating the level of inflammatory factor IL-6.
