摘要
目的探讨miR-151a-3p在胰腺癌细胞中的表达及miR-151a-3p表达对胰腺癌细胞增殖、迁移、侵袭及凋亡的影响。方法采用实时荧光定量聚合酶链反应(qRT-PCR)检测miR-151a-3p在三种胰腺癌细胞系的表达情况;利用脂质体转染技术将miR-151a-3p mimic和mimic NC转染至胰腺癌BxPC-3细胞中,qRT-PCR检测转染后miR-151a-3p在BxPC-3细胞中的表达水平;CCK-8法检测转染后BxPC-3细胞的增殖能力;Transwell实验检测转染后BxPC-3细胞的迁移及侵袭能力;流式细胞术检测转染BxPC-3细胞的凋亡能力。结果qRT-PCR结果显示,3种胰腺癌细胞系中miR-151a-3p表达水平均显著升高(P<0.01),其中,BxPC-3细胞中miR-151a-3p表达水平最高;转染miR-151a-3p mimic的BxPC-3细胞中miR-151a-3p表达水平显著高于mimic NC和空白组(P<0.001);CCK-8实验结果显示,转染miR-151a-3p mimic的BxPC-3细胞增殖能力显著高于mimic NC和空白对照组(P<0.01);Transwell实验结果显示,转染miR-151a-3p mimic的BxPC-3细胞迁移及侵袭能力显著高于mimic NC和空白对照组(P<0.01);流式细胞术结果显示,转染miR-151a-3p mimic的BxPC-3细胞凋亡率显著低于mimic NC和空白对照组(P<0.001)。结论miR-151a-3p在胰腺癌细胞中高表达,过表达miR-151a-3p促进胰腺癌细胞增殖、迁移及侵袭,抑制细胞凋亡。
Objective To investigate the expression of miR-151a-3p in pancreatic cancer cells and the effect of miR-151a-3p on proliferation,migration,invasion and apoptosis of pancreatic cancer cells.Methods Real-time fluorescent quantitative polymerase chain reaction(qRT-PCR)was used to detect the expression of miR-151a-3p in three pancreatic cancer cell lines.miR-151a-3p mimic and mimic NC were transfected into pancreatic cancer BxPC-3 cells using liposome transfection,qRT-PCR was used to detect the expression of miR-151a-3p in BxPC-3 cells after transfection.CCK-8 assay was used to detect the proliferation ability of BxPC-3 cells after transfection;Transwell assay was used to detect the migration and invasion ability of BxPC-3 cells after transfection;and flow cytometry was used to detect the apoptosis ability of BxPC-3 cells.Results The results of qRT-PCR showed that the expression of miR-151a-3p was significantly higher in all three pancreatic cancer cell lines(P<0.01),among which,the expression of miR-151a-3p in BxPC-3 cells was the highest;the expression of miR-151a-3p in BxPC-3 cells transfected with miR-151a-3p mimic was significantly higher than that in mimic NC and blank group(P<0.001).The results of CCK-8 showed that the proliferation ability of BxPC-3 cells transfected with miR-151a-3p mimic was significantly higher than that of mimic NC and blank group(P<0.01).The results of Transwell experiment showed that the migration and invasion ability of BxPC-3 cells transfected with miR-151a-3p mimic was significantly higher than that of mimic NC and blank group(P<0.01).The results of flow cytometry showed that the apoptosis rate of BxPC-3 cells transfected with miR-151a-3p mimic was significantly lower than that of mimic NC and blank group(P<0.001).Conclusion miR-151a-3p is highly expressed in pancreatic cancer cells.Overexpression of miR-151a-3p promotes the proliferation,migration,invasion,and inhibits apoptosis of pancreatic cancer cells.
