秦巴硒菇提取物FA-2-b-β对人多发性骨髓瘤细胞凋亡的影响 被引量:1

摘要 目的探讨秦巴硒菇提取物FA-2-b-β对人骨髓瘤RPMI8226细胞凋亡的作用。方法利用MTT比色法检测不同药物浓度(0.6、1.2、1.8、2.4mg/mL)对RPMI8226细胞的抑制率;流式细胞仪检测细胞凋亡率;Western blotting检测BCL-2和BAX蛋白的表达。结果 FA-2-b-β对RPMI8226细胞的增殖具有明显抑制作用,呈时间和剂量依赖性。流式细胞术检测显示,不同浓度FA-2-b-β作用于RPMI8226细胞48 h时,随药物浓度增加凋亡率也增高。Western blotting检测显示,凋亡相关蛋白BAX表达上调,BCL-2表达下调。结论 FA-2-b-β能够抑制多发性骨髓瘤细胞增殖及诱导其凋亡,其机制可能与BCL-2/BAX表达调控有关。 Objective To investigate the apoptosis mechanism of human multiple myeloma cell RPMI 8226 induced by Qinba selenium-mushroom extract(FA-2-b-β).Methods The proliferation ability of RPMI 8226 cells treated by various concentration of FA-2-b-β(0.6,1.2,1.8,2.4 mg/mL) in vitro for 24,48 and 72 hours were tested by MTT method. Flow cytometry was used to detect the apoptosis rate of RPMI 8226 in each group after treated by FA-2-b-β. Expression of BAX and BCL-2 protein were examined by Western blot.Result The proliferation of RPMI 8226 cells was inhibited significantly by FA-2-b-β,which showed a time-and dose-dependent manner. Flow cytometry showed that with the increasing of drug concentration,the apoptosis was also increased,when RPMI 8226 cells was treated by FA-2-b-β for 48,western blot indicated that the expression of apoptosis related protein BAX were upregulated,the expression of BCL2 were downregulated.Conclusion FA-2-b-β can inhibit the proliferative ability of RPMI 8226 cells,this mechanis may be associated with the downregulation of bcl-2 expression and increase the Bax expression.
