紫檀芪对异丙肾上腺素致心肌纤维化的作用研究 被引量:2

摘要 目的探讨紫檀芪对异丙肾上腺素(ISO)诱导的小鼠心肌纤维化的作用。方法选取ICR小鼠18只,随机分为正常对照组(皮下注射生理盐水7 mg/kg)、模型组(皮下注射异丙基肾上腺7 mg/kg)与治疗组(模型组基础上腹腔注射紫檀芪10 mg/kg)。根据心体比和Masson染色评价小鼠心脏功能和结构的改变,实时定量荧光PCR(qRT-PCR)检测心肌组织COL13A1的mRNA表达情况;酶联免疫吸附试验(ELISA)法检测血清和心肌组织中脑钠素(BNP)、羟脯氨酸(Hydro)的水平。结果模型组小鼠心脏心体比显著增大,治疗组小鼠心体比降低,差异有统计学意义(P<0.05);Masson染色显示模型组小鼠心肌发生纤维化,治疗组小鼠心肌纤维化有所改善;模型组心肌组织COL13A1 mRNA表达升高,治疗组小鼠表达降低,差异有统计学意义(P<0.05);模型组小鼠血清和心肌组织中的BNP及Hydro的含量均明显增加,差异有统计学意义(P<0.05);治疗组血清和心肌组织中BNP含量降低,差异均有统计学意义(P<0.05),且血清和心肌组织中的Hydro含量也降低,差异均有统计学意义(P<0.05)。结论紫檀芪可改善ISO诱导的小鼠心肌纤维化。 Objective To investigate the effect of pterostilbene on myocardial fibrosis induced by isoproterenol(ISO)in mice.Methods Eighteen ICR mice were randomly divided into negative control group(subcutaneous injection of normal saline 7 mg/kg),model group(subcutaneous injection of isoproterenol 7 mg/kg)and treatment group(model group was intraperitoneally injected with pterostilbene 10 mg/kg).Heart-to-body ratio was measured to evaluate the cardiac function in mice and Masson staining was used to observe the morphological changes of mouse myocardium.Real-time quantitative fluorescent PCR(qRT-PCR)was used to detect the expression of COL13A1 mRNA in myocardial tissue.Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of brain natriuretic peptide(BNP)and hydroxyproline(Hydro)in serum and myocardial tissue.Results Compared with the normal control group,the heart-to-body ratio of the model group was significantly increased,the difference was statistically significant(P<0.05).Masson staining showed the myocardial fibrosis.The expression of COL13A1mRNA was elevated in myocardial tissue.The difference was statistically significant(P<0.05).The levels of BNP and Hydro in serum and myocardial tissue were significantly increased,and the difference was statistically significant(P<0.05).Compared with the model group,the heart-to-body ratio of the rats in the group was significantly lower(P<0.05).The level of myocardial fibrosis was also significantly decreased and the expression of COL13A1mRNA was also decreased and the difference was statistically significant(P<0.05),the BNP content in serum and myocardial tissue decreased,the difference was statistically significant(P<0.05),and the Hydro content in serum and myocardial tissue also decreased,the difference was statistically significant(P<0.05).Conclusion Pterostilbene can improve ISO-induced myocardial fibrosis in mice.
