响应面法优化锁阳水提取物提取工艺

摘要 在单因素试验基础上,应用Box-Behnken中心组合设计原理,以提取时间、提取次数、液料比为自变量,锁阳水提取物中没食子酸、原儿茶酸含量为响应值,设计3因素3水平响应面分析试验,确定锁阳水提取物的最佳提取工艺为:液料比30∶1(mL/g);提取时间45min;提取2次,各因素的影响作用依次为提取次数>液料比>提取时间.锁阳水提取物中没食子酸、原儿茶酸含量预测值分别为0.992mg/g、0.189mg/g,实际值分别为0.986mg/g、0.185mg/g,相对偏差分别为0.61%和2.70%,证明模型理论预测值与实际值拟合效果良好.因此利用响应面法得到的锁阳水提取物提取工艺准确可靠,预测性良好. On the basis of single factor experiment,Box-Behnken central composite design principle was applied to determine the optimum extraction process of Cynomorium Songaricum Rupr. water extraction,taking extraction time,extraction times and liquid-to-material ratio as independent variables,and the content of gallic acid and protocatechuic acid in Cynomorium songaricum Rupr. water extraction as response values. The optimum extraction technology of Cynomorium songaricum Rupr. water extract was as follows:the ratio of liquid to material was 30:1(mL/g);the extraction time was 45 minutes;the extraction time was 2 times,and the effect of each factor was extraction times >liquid to material ratio > extraction time in turn. The predicted values of gallic acid and protocatechuic acid in Cynomorium songaricum Rupr. water extract were 0.992 mg/g and 0.189 mg/g respectively. The actual values were 0.986 mg/g and 0.185 mg/g,and the relative deviations were 0.61% and 2.70% respectively. It proved that the theoretical predicted values of the model were in good agreement with the actual values. The extraction process of Cynomorium songaricum Rupr. water extraction obtained by response surface methodology is accurate,reliable and predictable.
