葛根素对MCF-7细胞的雌激素样促增殖作用分析

目的对葛根素对MCF-7细胞的雌激素样促增殖作用加以分析。方法 2018年1月-2019年6月间选取15例乳腺癌患者展开研究,提取细胞。将人体MCF-7细胞作为靶细胞,将雌酚酮1×10^-8mol/L设定为阳性对照组,并设定空白对照组,将葛根干预组依据浓度差异分别设定为1×10^-8mol/L组、1×10^-7mol/L组及1×10^-6mol/L组。①将细胞浓度调整为4×10^4个/mL,以12孔培养板接种,48 h后,借助倒置显微镜对细胞形态予以观察;②将细胞浓度调整为2×10^4个/L,以96孔培养板接种,以四唑盐法对平均增值率加以计算;③将细胞浓度控制于0.9×10^4个/mL,以24孔培养板接种,经染色后,借助光镜对活细胞数加以计量,取均值,绘制细胞生长曲线。对葛根素对MCF-7细胞雌激素样促增殖影响加以分析。结果据数据分析,经葛根素1×10^-7mol/L及1×10^-6mol/L处置后第1天、第2天及第3天同对照相较而言,增殖速度均明显加快。第1天、第2天及第3天,组间差异有统计学意义(t=3.955、4.012、3.946、3.867、4.002,P<0.001)。结论葛根素可加快MCF-7细胞增殖,表明具备弱雌激素样作用。

Objective To analyze the estrogen-like pro-proliferation effect of gegensin to MCF-7 cells. Methods From January 2018 to June 2019, 15 breast cancer patients were selected for research and cell extraction.The human MCF-7 cells were targeted, the cents of estrogen1 1 ×10^-8 mol/L were set as a positive control group, and the blank control group was set, and the Gegen intervention group was set to 1×10^-8 mol/L group according to concentration difference, 1×10^-7 mol/L group and 1×10^-6 mol/L group. 1. adjusts the cell concentration to 4 ×10^4 pcs/mL, inoculates with 12-well culture plate, 48 h, the cell morphology is observed with the help of an inverted microscope.2. the cell concentration is adjusted to 2×10^4 pcs/L, the 96-well culture plate is inoculated, the average value-added rate is calculated by the tetrazolian salt method.3. the cell concentration is controlled at 0.9×10^4 pcs/mL, inoculated with a 24-well culture plate,after staining, with the help of a mirror to measure the number of living cells, take the mean, draw the cell growth curve. The effect son of gegenc on estrogen-like pro-proliferation in MCF-7 cells was analyzed. Results According to the data analysis, the proliferation rate of puerarin treated at 1×10^-7 mol/L and 1×10^-6 mol/L on the 1 st, 2 nd and 3 d was significantly faster than that of the control. At 1 d, 2 d, and 3 d, the, respectively, and the difference between groups were statistically significant(t=3.955, 4.012, 3.946, 3.867, 4.002,P<0.001). Conclusion Gergensin can accelerate THE proliferation of MCF-7 cells, indicating that it has a weak estrogen-like effect.
