摘要
目的:探讨独一味胶囊联合维素B12(VB12)治疗复发性口腔溃疡大鼠的效果。方法:从72只SPF级SD大鼠中随机抽取12只制备免疫法建模的乳化剂。剩余60只大鼠随机分为正常组、模型组、醋酸泼尼松龙注射液组、独一味胶囊组、独一味胶囊联合VB12组。模型组、醋酸泼尼松龙注射液组、独一味胶囊组、独一味胶囊联合VB12组采用免疫法建立复发性口腔溃疡模型。造模成功后各组大鼠分别灌胃给药治疗20 d后,观察各组大鼠临床症状的改善情况(溃疡数目、持续时间和间隔时间);利用Western blot检测各组大鼠中核因子κB p65(NF-κB p65)、肿瘤坏死因子α(TNF-α)及内皮生长因子(VEGF)表达水平;比较各组大鼠血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSHPx)及丙二醛(MDA)水平。结果:独一味胶囊联合VB12组大鼠溃疡数目及持续时间均少于模型组与醋酸泼尼松龙注射液组、独一味胶囊组,间隔时间均长于模型组与醋酸泼尼松龙注射液组、独一味胶囊组(P<0.05);醋酸泼尼松龙注射液组、独一味胶囊组、独一味胶囊联合VB12组大鼠NF-κB p65、TNF-α、MDA水平均明显低于模型组,VEGF、SOD及GSHPx水平明显高于模型组,且独一味胶囊联合VB12组大鼠中上述指标水平变化趋势明显高于醋酸泼尼松龙注射液组、独一味胶囊组(P<0.05)。结论:独一味胶囊联合VB12可显著减少大鼠口腔溃疡数目,缩短愈合时间,其作用机制可能与下调黏膜组织中炎症因子表达,缓解氧化损伤有关。
Objective:To investigate the effect of Duyiwei capsule combined with VB12in the treatment of recurrent oral ulcer rats.Methods:Totally 12 SD rats were randomly selected from 72 SPF-class SD rats,to prepare immunoassay-modeled emulsifiers.The remaining 60 rats were randomly divided into normal group,model group,Prednisolone acetate ijection group,Duyiwei capsule group and Duyiwei capsule combined with VB12group.The model group,Prednisolone acetate ijection group,Duyiwei capsule group and Duyiwei capsule combined with VB12group were established the model of recurrent aphthous ulcer by immune method.The rats in each group were administrated by gavage for 20 days after the model was established successfully.The improvement of clinical symptoms(number of ulcers,duration and interval time)was observed.The expression levels of NF-κB p65,TNF-αand vascular endothelial growth factor(VEGF)in rats of each group were detected by Western blot.The levels of superoxide dismutase(SOD),glutathione peroxidase(GSHPx)and malondialdehyde(MDA)in rats of each group were compared.Results:The Duyiwei capsule combined with VB12group showed less quantity and duration of ulcer,as well as longer interval time than model group,Prednisolone acetate ijection group and Duyiwei capsule group(P<0.05).Prednisolone acetate ijection group,Duyiwei capsule group and Duyiwei capsule combined with VB12group showed lower levels of NF-κB p65,TNF-αand MDA than model group,while higher levels of VEGF,SOD and GSHPx than model group,and the change trend of the above indexes in the Duyiwei capsule combined with VB12group was significantly greater than that in the Prednisolone acetate ijection group and Duyiwei capsule group(P<0.05).Conclusion:Duyiwei capsule combined with VB12can significantly reduce the number of oral ulcers in rats and shorten the healing time.Its mechanism may be related to down-regulating the expression of inflammatory factors in mucosal tissue and alleviating oxidative damage.
