摘要
目的建立反相高效液相色谱(RP-HPLC)法同时测定文冠木中表没食子儿茶素、儿茶素、表儿茶素、二氢杨梅素、二氢槲皮素、杨梅素6个成分含量的方法。方法采用岛津Inertsil C18色谱柱(250 mm×4.6 mm,5.0μm);乙腈(A)-0.4%乙酸水(B)为流动相,梯度洗脱;流速:1.0 mL·min-1;柱温:20℃;检测波长为280 nm。结果表没食子儿茶素、儿茶素、表儿茶素、二氢杨梅素、二氢槲皮素、杨梅素的进样量分别在0.170~4.080μg(r=0.9992)、0.070~0.840μg(r=0.9996)、0.200~2.400μg(r=0.9997)、0.270~3.240μg(r=1.0000)、0.032~0.384μg(r=1.0000)、0.073~0.876μg(r=1.0000)范围内线性关系良好;平均回收率(n=3)为96.62%~101.28%,RSD为1.1%~2.5%。9批文冠木药材中表没食子儿茶素、儿茶素、表儿茶素、二氢杨梅素、二氢槲皮素、杨梅素的含量分别为1.78~7.26 mg·g^-1、0.11~1.31 mg·g^-1、1.34~4.52 mg·g^-1、1.07~5.33 mg·g^-1、0.02~0.85 mg·g^-1、0.05~1.45 mg·g^-1。结论所建立的HPLC方法专属性强、准确,重复性良好,可为文冠木药材的质量控制提供科学依据。
Objective To establish a RP-HPLC method for simultaneous determination of epigallocatechin,catechin,epicatechin,dihydromyricetin,dihydroquercetin,and myketin in Xanthocerais lignum. Methods The analysis was performed at 20 ℃ on an Inertsil C18 column(250 mm ×4.6 mm,5.0 μm),eluted with a gradient program using acetonitrile(A)-0.4% acetic acid aqueous solution(B) as the mobile phase to determine the contents of the six components. The flow rate was 1.0 mL·min-1,and the detection wavelength was 280 nm. Results The linear ranges of six components were 0.170-4.080 μg(r=0.999 2)for epigallocatechin,0.070-0.840 μg(r=0.999 6)for catechin,0.200-2.400 μg(r=0.999 7)for epicatechin,0.270-3.240 μg(r=1.000 0)for dihydromyricetin,0.032-0.384 μg(r=1.000 0)for dihydroquercetin,and 0.073-0.876 μg(r=1.000 0)for myketin. The average recoveries(n=3)of six components were between 96.62%-101.28% with RSD of 1.1%-2.5%. The contents of six components in nine samples were 1.78-7.26 mg·g^-1 for epigallocatechin,0.11-1.31 mg·g^-1 for catechin,1.34-4.52 mg·g^-1 for epicatechin,1.07-5.33 mg·g^-1 for dihydromyricetin,0.02-0.85 mg·g^-1 for dihydroquercetin,and 0.05-1.45 mg·g^-1 for myketin.Conclusion The method established in this research for determination of the six components is specific,simple,accurate,reproducible,which can provide a scientific basis for quality control of Xanthocerais lignum.
