丹参水提取液在小鼠体内抑菌效果及大肠杆菌感染的预防作用 被引量:3

摘要 【目的】通过对小鼠灌服一段时间的丹参水提取液,研究丹参对小鼠的体内抑菌效果,丹参水提取液对作用于大肠杆菌O101的小鼠的预防效果,从而探讨丹参在体内的抗菌作用。【方法】用试验动物小鼠验证丹参的体内抑菌作用,对灌服不同时间段丹参水提取液小鼠的肠道内容物进行细菌计数,同时观察小鼠体质量增加、脾脏指数、肝脏指数等指标与灌服丹参水提取液之间的关系;同时通过对饲喂一段时间丹参水提取液的小鼠进行大肠杆菌O101攻毒试验,进行体内抑菌试验,通过观察小鼠的存活率以及攻毒后肠道菌群计数判定丹参水提取液的体内抑菌效果。【结果】丹参水提取液提高了小鼠的生长性能,中剂量组、高剂量组体质量增加效果差异极显著(P<0.001);中剂量组、高剂量组对乳酸杆菌的增菌效果差异显著(0.001<P<0.01)或极显著(P<0.001);高剂量组对金黄色葡萄球菌抑菌作用极显著(P<0.001)、高剂量组对大肠杆菌抑菌作用极显著(P<0.001);对大肠杆菌感染具有防护作用。对饲喂28 d丹参水提取液的小鼠进行大肠杆菌O101攻毒,空白组死亡6只,低剂量组死亡4只,中剂量组死亡3只,高剂量组死亡4只。【结论】丹参水提取液有增加肠道内乳酸杆菌数量并减少大肠杆菌和金黄色葡萄球菌数量的趋势,对其成长性能,免疫器官发育都有促进作用。攻毒后小鼠的盲肠内容物菌群分别有促进或抑制的作用,对益生菌起到促进作用,对病原菌起到抑制的作用,同时减少小鼠死亡数量。 【Objective】The antibacterial effect of S.miltiorrhiza Bunge on mice was studied by administering the S.miltiorrhiza water extract,and the preventive effect of S.miltiorrhiza extract on mice against E.coli O 101 was studied to explore the effect of S.miltiorrhiza in vivo Antibacterial effect.【Method】The bacteriostasis of S.miltiorrhiza was verified in mice.The intestinal contents of the mice were filled with S.miltiorrhiza water extract at different time points,and the bacterial counts of the mice were compared.At the same time,the relationship between body weight gain,spleen index,liver index and the S.miltiorrhiza water extract was observed.The experiment was carried out by E.coli O 101 challenge test on mice fed with S.miltiorrhiza water extract.The antibacterial effect in vivo was determined by observing the survival rate of the mice and the count of intestinal flora after challenge.【Result】The water extract of S.miltiorrhiza increased the growth performance of mice.The difference of weight gain between the middle and high dose groups was extremely significant(P<0.001).The effect of the medium and high dose groups on the growth of Lactobacillus was significant(0.001<P<0.01).Or extremely significant(P<0.001);the high-dose group had a significant antibacterial activity against Staphylococcus aureus(P<0.001),and the high-dose group had a significant antibacterial effect on E.coli O 101;Protective effect.E.coli O 101 was challenged in mice fed with water extract of S.miltiorrhiza for 28 days.Six mice died in the blank group,4 died in the low dose group,3 died in the middle dose group,and 4 died in the high dose group.【Conclusion】S.miltiorrhiza water extract has a tendency to increase the number of lactobacilli in the intestine and reduce the number of E.coli and staphylococcus aureus,and promote its growth performance and intestinal development.The S.miltiorrhiza water extracthas an inhibitory effect on E.coli and reduces deaths in mice.
