FOXO1基因在妊娠糖尿病血中的表达及与胰岛素的相关性

Expression of FoxO1 gene and its relationship with insulin in blood of gestational diabetes mellitus

目的探究妊娠糖尿病(GDM)血中FOXO1基因的表达情况,及其与胰岛素的相关性研究。方法采集来贵州中医药大学第二附属医院产科门诊进行产检的GDM孕妇和正常孕妇外周血108例,分别作为GDM研究组和正常对照组,用葡萄糖氧化酶法测定血糖,电化学发光法测定胰岛素,ELISA法检测FOXO1的蛋白表达,Real-time PCR检测FOXO1 mRNA的表达,应用稳态模式评估法计算胰岛素抵抗指数(HOMA-IR),分析孕妇外周血FOXO1基因的表达与HOMA-IR的相关性。结果与正常孕妇组相比,GDM孕妇研究组外周血胰岛素水平、FOXO1 mRNA和蛋白的表达均高于正常孕妇组(P<0.05);Pearson分析显示FOXO1的mRNA和蛋白的表达均与HOMA-IR呈正相关(r=0.555,r=0.530,P<0.05)。结论FOXO1可能参与了GDM的生理病理过程;且其在外周血中的表达与GDM孕妇胰岛素抵抗呈正相关,可为GDM的血清学诊断提供新的理论依据和研究方向。

Objective To investigate the expression of FoxO1 gene in blood of gestational diabetes mellitus(GDM)and its correlation with insulin.Methods The peripheral blood of 108 GDM pregnant women and normal pregnant women were collected from the obstetric clinic of the Second Affiliated Hospital of Guizhou University of Traditional Chinese Medicine(TCM).They were used as GDM study group and normal control group,respectively.Glucose was measured by glucose oxidase method,insulin was measured by electrochemiluminescence method,FoxO1 protein expression was detected by ELISA method,FoxO1mRNA expression was detected by real-time PCR.The relationship between the expression of FoxO1 gene and HOMA-IR was analyzed by the steady-state mode evaluation method.Results Compared with normal pregnant women,the expression of insulin,FoxO1mRNA and protein in peripheral blood of GDM pregnant women group was higher than that of normal pregnant women group(P<0.05).Pearson analysis showed that the expression of FoxO1mRNA and protein was positively correlated with HOMA-IR(r=0.555,r=0.530,P<0.05).Conclusion FoxO1 may participate in the physiological and pathological process of GDM,and its expression in the peripheral blood is positively correlated with the insulin resistance of GDM pregnant women,which can provide a new theoretical basis and research direction for the serodiagnosis of GDM.
