消炎退热合剂对脂多糖致急性肺损伤模型小鼠炎症因子表达的影响 被引量:1

Effect of Xiaoyan Tuire Mixture on the Expression of Inflammatory Factors in Lipopolysaccharide Induced Acute Lung Injury Model Mice

摘要 目的:观察消炎退热合剂对脂多糖(lipopolysaccharide,LPS)诱导的肺损伤模型小鼠肺组织中炎性因子表达的影响。方法:取ICR小鼠,采用多次腹腔注射LPS建立小鼠肺损伤模型,随机分为模型组,地塞米松组(2 mg·kg^-1),消炎退热合剂小剂量组(3 g·kg^-1)、中剂量组(6 g·kg^-1)、大剂量组(12 g·kg^-1)。各组灌胃给药,连续4周。末次给药后2 h采血,测定血清白细胞介素(interleukin,IL)-4、IL-12、IL-10、干扰素-γ(interferon-γ,IFN-γ)、IL-6、肿瘤坏死因子-α(tumor necrosis factor,TNF-α)水平;PBS磷酸缓冲液进行肺灌洗,检测灌洗液中的IL-6和TNF-α水平。取左肺组织,常规石蜡切片,HE染色,光镜下观察肺组织病理学特征。结果:消炎退热合剂中剂量组、大剂量组右上叶组织湿质量/干质量显著低于模型组(P<0.01)。与模型组比较,消炎退热合剂小剂量组、中剂量组、大剂量组的IL-12、IFN-γ、IL-10和IL-4水平均显著降低(P<0.05或P<0.01);肺泡灌洗液中,消炎退热合剂小剂量组、中剂量组、大剂量组的IL-6水平明显降低(P<0.01),消炎退热合剂大剂量组中TNF-α水平明显降低(P<0.05)。HE染色结果显示,与模型组比较,消炎退热合剂大剂量组小鼠肺泡间隔仅局部有轻度增厚,轻度炎细胞浸润,无出血。结论:消炎退热合剂能有效降低LPS诱导的肺损伤模型小鼠的炎症水平。 Objective:To observe the effect of Xiaoyan Tuire Mixture on the expression of inflammatory factors in the lung tissue of model mice induced by lipopolysaccharide(LPS).Methods:Take ICR mice and use multiple intraperitoneal injections of LPS to establish a mouse lung injury model,randomly divided into a model group,a dexamethasone group(2 mg·kg^-1),and a small dose group(3 g·kg^-1),a medium dose group(6 g·kg^-1),a large dose group(12 g·kg^-1)of Xiaoyan Tuire Mixture.Each group was given by intragastric administration for 4 consecutive weeks.The serum levels of interleukin-4(IL-4),IL-12,IL-10,interferon-γ(IFN-γ),IL-6,tumor necrosis factor-α(TNF-α)were measured 2 hours after the last administration.The level of IL-6 and TNF-αin PBS phosphate buffer was detected.The left lung tissue was taken and paraffin section was made.HE staining was used to observe the histopathological characteristics of the lung.Results:The wet mass/dry mass of the right upper lobe in the middle dose group and the high dose group of Xiaoyan Tuire Mixture were significantly lower than that in the model group(P<0.01).Compared with the model group,the levels of IL-12,IFN-γ,IL-10 and IL-4 in the small dose group,the middle dose group and the large dose group of Xiaoyan Tuire Mixture decreased significantly(P<0.05 or P<0.01).In BALF,the level of IL-6 in low dose group,middle dose group and high dose group decreased significantly(P<0.01),and the level of TNF-αin high dose group decreased significantly(P<0.05).The results of HE staining showed that,compared with the model group,the alveolar septa of mice in the high dose group of Xiaoyan Tuire Mixture only had slight thickening,mild inflammatory cell infiltration and no bleeding.Conclusion:Xiaoyan Tuire Mixture can effectively reduce the inflammatory level of LPS induced lung injury model mice.
