NEAT1 Inhibits Osteoporosis through the PI3K/AKT/Bcl-2 Signaling Pathway

摘要 为探讨NEAT1在骨质疏松症中的作用以及可能的病理机制,本研究通过建立卵巢去势和鼠尾悬挂2种骨质疏松的小鼠模型,将C57BL/6分为假手术组(Sham组)、OVX组和TS组;经过PCR测定小鼠NEAT1的表达;Elisa法检测小鼠E2、ALP和TRACP水平;Western blotting检测细胞凋亡因子PI3K/AKT/Bcl-2的蛋白水平。结果显示,建模4周后,3组小鼠体重没有显著变化;与Sham组相比,OVX组和TS组小鼠的骨密度值显著降低,骨生化指标ALP和TRACR水平明显升高;OVX组小鼠的E2水平与Sham组相比明显降低;与Sham组相比,OVX组和TS组小鼠的NEAT1表达显著下调;与Sham组相比,OVX组和TS组小鼠p-AKT和Bcl-2蛋白水平明显降低。本研究结果表明,NEAT1可能通过抑制PI3K/AKT/Bcl-2细胞凋亡途径诱导骨质疏松。 To investigate the role of NEAT1 in osteoporosis and its possible pathological mechanism,in this study,C57 BL/6 was divided into Sham group,OVX group and TS group by establishing two kinds of mouse models of osteoporosis including ovarian castration and rat tail suspension.The expression of NEAT1 was detected by PCR.E2 method was used to detect mouse E2 and ALP,and TRACP levels.Western blotting analysis of the apoptotic factor PI3 K/AKT/Bcl-2 protein levels.After four weeks of modeling,the results showed that there was no significant change in body weight of the three groups of mice.Compared with the Sham group,the bone mineral density values of the OVX group and the TS group were significantly decreased,and the bone biochemical indicators ALP and TRACT were significantly increased.The E2 level of the rats was significantly lower than that of the sham group;compared with the Sham group,the NEAT1 expression was significantly down-regulated in the OVX group and the TS group;Compared with the Sham group,the OVX group and the TS group were p-AKT and Bcl-2 protein levels were significantly reduced.The results of this study suggested that NEAT1 might induce osteoporosis by inhibiting the apoptosis pathway of PI3 K/AKT/Bcl-2 cells.
