珠芽景天不同部位提取物抗小鼠急性肝损伤的作用研究 被引量:2

摘要 目的筛选珠芽景天具有抗肝损伤作用的活性部位。方法采用D101大孔吸附树脂法对珠芽景天70%乙醇提取物进行分段富集,依次洗脱得到珠芽景天的水、50%乙醇、95%乙醇洗脱部位。将120只KM小鼠随机分为正常对照组(0.5%羧甲基纤维素钠20 mL/kg)、模型组(0.5%羧甲基纤维素钠20 mL/kg)、双环醇组(阳性对照组,0.2 g/kg),以及水段高、中、低剂量组(以生药量计分别为16.38,10.92,5.46 g/kg),50%乙醇段高、中、低剂量组(以生药量计分别为10.08,6.72,3.36 g/kg),95%乙醇段高、中、低剂量组(以生药量计分别为2.46,1.64,0.82 g/kg),每组10只,每天1次,灌胃7 d。末次给药2 h后,正常对照组腹腔注射玉米油10 mL/kg,其余各组腹腔注射0.1%四氯化碳(CCl4)-玉米油混合液,成模16 h后摘取小鼠眼球取血,分离小鼠肝脏组织。测定不同组小鼠血清中丙氨酸氨基转移酶(ALT)、天门冬酸氨基转移酶(AST)活性与肝匀浆中丙二醛(MDA)含量,观察其肝组织病理形态学变化。结果与正常对照组比较,模型组小鼠血清中ALT和AST活性显著升高,肝组织中MDA含量显著升高(P<0.01);与模型组比较,双环醇组和50%乙醇段高、中剂量组小鼠血清中ALT和AST的活性均显著降低(P<0.05),肝组织中MDA含量显著降低(P<0.05)。结合病理学切片观察,50%乙醇段高、中、低剂量组小鼠急性肝损伤均有不同程度减轻。结论珠芽景天70%乙醇提取物的50%乙醇洗脱部位为其具有抗CCl4致小鼠急性肝损伤作用的活性部位。 Objective To investigate the active part of Sedum bulbiferum against acute liver injury.Methods The 70%ethanol extract of Sedum bulbiferum was segmented and enriched by D101 macroporous adsorption resin to obtain the elution of water,50%ethanol and 95%ethanol.A total of 120 KM mice were randomly divided into normal control group,model group,bicyclol group(positive control,0.2 g/kg),high-dose,medium-dose,low-dose groups(16.38,10.92,5.46 g/kg,calculated by crude drug)of water extract,high-dose,medium-dose,low-dose groups(10.08,6.72,3.36 g/kg,calculated by crude drug)of 50%ethanol extract,high-dose,medium-dose,l ow-dose groups(2.46,1.64,0.82 g/kg,calculated by crude drug)of 95%ethanol extract,10 mice in each group.The normal control group and the model group were given 0.5%CMC-Na,and the other groups were given the corresponding drugs intragastrically once a day for consecutive 7 d.Two hours after the last medication,the normal control group were given corn oil(10 mL/kg)by intraperitoneal injection,while the mice in other groups were given 0.1%CCl4 corn oil solution to set up acute liver injury model.The mice's eyeballs and blood were taken,and the liver tissue of the mice were isolated after 16 h.The serum contents of alanine aminotransferase(ALT),aspartate aminotransferase(AST)and malondialdehyde(MDA)in different groups of mice were determined.The liver histopathological changes were observed.Results Compared with the normal control group,t he activity of ALT and AST in the serum of the model group was significantly increased,and the MDA content in the liver tissue was significantly increased(P<0.01);compared with the model group,the activity of ALT and AST in the serum of mice was significantly reduced in the bicyclol group and the high-dose group and the medium-dose group of 50%ethanol extract(P<0.05),and the content of MDA in liver tissue was significantly reduced(P<0.05).The pathological sections showed that liver injury was relieved to a certain extent in high-dose,medium-dose,low-dose group of 50%ethanol extract.Conclusion The 50%ethanol segment of the 70%ethanol extract from Sedum bulbiferum is the active part of anti-acute liver injury induced by CCl4 in mice.
