摘要
目的黄芩茎叶总黄酮(SSTF)应用于高甘油三酯血症方面的研究很少,文中以高甘油三酯血症模型大鼠为对象,探讨黄芩茎叶总黄酮对高甘油三酯血症大鼠脂代谢及氧化应激水平的影响。方法60只SD大鼠随机数字表法分为正常组、模型组、非诺贝特(100 mg/kg)组以及SSTF低(50 mg/kg)、中(100 mg/kg)、高(200 mg/kg)剂量组。除正常组外,其他5组均喂食高脂饲料建立高甘油三酯血症大鼠模型,并同时给予相应药物干预6周,测量大鼠体重、肝湿重计算肝指数,测定血清TG、TC、HDL-C、LDL-C,肝组织匀浆测定TG、TC、丙二醇、SOD、GSH-PX,HE染色观察肝组织病理形态学变化。结果与正常组比较,模型组大鼠2、4、6周体重、6周肝指数增加(P<0.05),血清TG、TC、LDL-C水平升高(P<0.05),HDL-C水平降低(P<0.05),模型组大鼠肝组织TG[(1.60±0.15)mmol/L]较正常组[(0.55±0.12)mmol/L]升高(P<0.05)、TC[(3.47±0.36)mmol/L]水平较正常组[(1.36±0.18)mmol/L]升高(P<0.05),丙二醇[(10.29±1.98)nmol/mg]较正常组[(4.81±1.12)nmol/mg]表达增加(P<0.05);SOD[(106.84±24.01)U/mg]、GSH-PX[(9.28±2.08)U/mg]较正常组[(206.72±29.40)、(15.58±1.56)U/mg]表达降低(P<0.05)。与模型组比较,非诺贝特组、SST各剂量组大鼠2、4、6周体重及6周肝指数减少(P<0.05),血清TG、TC、LDL-C水平降低(P<0.05),HDL-C水平升高(P<0.05),肝组织TG、TC水平降低(P<0.05),丙二醇表达减少(P<0.05),SOD、GSH-PX表达增加(P<0.05)。与非诺贝特组相比,SSTF高剂量组大鼠4、6周体重降低(P<0.05)、6周肝指数减少(P<0.05),血清TG、LDL-C水平降低(P<0.05),HDL-C水平升高(P<0.05);SSTF高剂量组肝组织TG[(0.72±0.14)mmol/L]水平较非诺贝特组(0.86±0.14)mmol/L降低(P<0.05),丙二醇[(5.07±1.31)nmol/mg]较非诺贝特组[(6.21±1.06)nmol/mg]表达减少(P<0.05),SOD[(172.21±30.21)U/mg]、GSH-PX[(14.62±2.42)U/mg]表达较非诺贝特组[(142.45±32.02)、(12.52±1.74)U/mg]增加(P<0.05)。结论黄芩茎叶总黄酮具有调节高甘油三酯血症大鼠脂代谢,改善肝病理性损伤的作用,该作用可能与抑制脂质过氧化、减少氧化应激有关。
Objective The study of the treatment of hypertriglyceridemia with scutellaria baicalensis stem-leaf total flavonoid(SSTF)are rarely reported.The objective of this study is to investigate the effects of SSTF on lipid metabolism and oxidative stress in rats with hypertriglyceridemia.Methods 60 SD rats were randomly divided into normal group,model group,fenofibrate group(100 mg·kg^-1),SSTF groups with low,medium and high doses(50,100,200 mg·kg^-1,respectively).All rats,except those of the normal group,were fed with high-fat diet and given corresponding drug intervention for 6 weeks.Then the body masses at the 2^nd,4^th and 6^th weeks as well as wet weight of the liver at the end of 6 th week were recorded,and liver index was calculated.The serum levels of triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)were detected.The levels of TG,TC,malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)in liver tissue were determined,and liver histopathological changes were observed by hematoxylin-eosin(HE)staining.Results In the model group,compared with the normal group,the body masses at the 2^nd,4^th and 6^th weeks and liver index at the end of 6^th week were increased,the serum levels of TG,TC and LDL-C got increased with decreased HDL-C level,and the levels of TG[(1.603±0.146)mmol/L],TC[(3.474±0.356)mmol/L]and MDA[(10.288±1.979)nmol/mg]in liver tissue were increased with decreased levels of SOD[(106.840±24.014)U/mg]and GSH-PX[(9.278±2.079)U/mg].Compared with the model group,the fenofibrate group and all SSTF groups showed decreased body masses at the 2^nd,4^th and 6^th weeks and liver index at the end of 6^th week,decreased serum levels of TG,TC and LDL-C with increased level of HDL-C,and decreased levels of TG,TC and MDA with increased levels of SOD and GSH-PX in liver tissue.The comparsion between the fenofibrate group and the high-dose SSTF group revealed that the body masses at the 4^th and 6^th weeks and liver index at the end of 6^th week were decreased,the serum levels of TG,TC and LDL-C were decreased with increased level of HDL-C,and the levels of TG[(0.718±0.135)mmol/L]and MDA[(5.071±1.305)nmol/mg]in liver tissue got decreased with increased levels of SOD[(172.210±30.214)U/mg]and GSH-PX[(14.623±2.418)U/mg]in the latter group.All the above-mentioned differences were statistically significant(all P<0.05).Conclusion SSTF can regulate lipid metabolism and improve pathological injury of liver in hypertriglyceridemia rats,which may be related with inhibition of lipid peroxidation and reduction of oxidative stress.
