HPLC-MS/MS同时检测血液中美利曲辛和氟哌噻吨

摘要 目的建立HPLC-MS/MS分析血液中美利曲辛和氟哌噻吨的方法。方法血液用甲醇/乙腈(1:9)提取后,用C18色谱柱进行分析,以水-甲醇(含0.1%甲酸)为流动相梯度洗脱,采用液相色谱-串联质谱SRM正离子模式检测。结果血中美利曲辛和氟哌噻吨的线性范围广,线性关系良好,相关系数r均大于0.999,美利曲辛和氟哌噻吨最低检测限分别为0.01ng/mL(S/N=8)、0.2ng/mL(S/N=12)。日内、日间精密度均小于10%。结论建立的方法准确度高、灵敏度好,可用于血液中美利曲辛和氟哌噻吨的同时定性、定量分析。 A HPLC-MS/MS method for the determination of melitracen and flupentixol in blood was established.After extracted with methanol/acetonitrile(1:9),they were analyzed by positive ion and SRM mode,ZORBAX Eclipse Plus C18 column.Water-methanol(containing 0.1%formic acid)was used as mobile phase gradient elution.The experimental results showed that melitracen and flupentixol in blood had a wide linear range and good linear relationship The correlation coefficient was greater than 0.999.The minimum detection limits of melitracen and flupentixol were 0.01ng/mL(S/N=8)and 0.2ng/mL(S/N=12),respectively.The intra-day and inter-day precision were both less than 10%.The method has high accuracy and good sensitivity.It can be used for simultaneous qualitative and quantitative detection of melitracen and flupentixol in blood.
