摘要
目的探讨黄芪发酵液对大鼠衰老模型的丙二醛(MDA)、脂褐素(LPF)、谷胱甘肽过氧化酶(GSH-Px)水平的影响。方法选自然衰老SD大鼠20~22月龄40只,随机的方法分为4组,即为老年空白对照组10只,发酵黄芪低剂量组10只,发酵黄芪中剂量组10只,发酵黄芪高剂量组10只;另选取2~3月龄10只作为青年空白对照组,高、中、低剂量组分别给予黄芪发酵液400、200、100 mg/kg·d,对照组灌胃等容积生理盐水,1次/d,连续进行8周。测定大鼠MDA、LPF、GSH-Px水平以及p16基因。结果高、中、低剂量组的大鼠体重均较青年空白对照组和老年空白对照组显著增加,且老年空白对照组较青年空白对照组有显著增高,差异有统计学意义(P<0.05)。与青年空白对照组比较,老年空白对照组的脑、心中的MDA、LPF的指标水平显著升高,差异有统计学意义(P<0.05);与老年空白对照组比较,高、中、低剂量组的MDA、LPF指标水平显著下降,差异有统计学意义(P<0.05)。与青年空白对照组比较,老年空白对照组的GSH-Px的指标水平显著降低,差异有统计学意义(P<0.05),与老年空白对照组比较,高、中、低剂量组均显著升高,差异有统计学意义(P<0.05)。进一步对高、中、低剂量进行组间比较,MDA、LPF指标水平高剂量组<中剂量组<低剂量组,GSH-Px的指标水平高剂量组>中剂量组>低剂量组,但差异无统计学意义(P>0.05)。老年空白对照组大鼠脾p16蛋白的表达显著高于青年空白对照组,差异有统计学意义(P<0.05)。免疫组织化学阳性结果显示细胞质呈现红棕或棕褐染色为阳性表达。结论黄芪发酵液能够显著降低大鼠衰老模型中MDA、LPF的生成,增高GSH-Px水平,从而发挥抗衰老作用,与其剂量无关。
Objective To investigate the effects of astragalus fermentation liquor on MDA,lipofuscin and GSH-Px levels in aging rats.Methods Selected natural aging SD rats 20~40 which was 22 months old,in addition to 10 mice 2~3 months old as young control grop.And the aging SD rats were were randomly divided into four groups;senile blank control group,low dose group,midum dose group,high dose group,and the dosage of astragalus fermented liquidmg was separately given 100,200 and 400 mg in each group.The levels of MDA,lipofuscin,GSH-Px and p16 genes in rats were measured.Results The weight of rats in the high,medium and low dose groups was significantly increased compared with the control group and the elderly blank control group(P<0.05),but there was no significant difference among the three groups(P>0.05).Compared with the control group,the levels of MDA and LPF in the brain and heart of the elderly blank control group were significantly increased(P<0.05).Compared with the elderly blank control group,the levels of MDA and LPF in the high,medium and low dose groups decreased significantly(P<0.05).Compared with control group,the elderly of GSH-Px index of the blank control group were significantly decreased,the significant difference(P<0.05),compared with old blank control group,the levels of GSH-Px index in high,medium and low dose groups were all significantly increased(P<0.05).There was no significant difference of MDA and LPF among the three groups:high dose group,medium dose group and low dose group(P>0.05).The expression of p16 protein in the spleen of the elderly blank control group was significantly higher than that of the young blank control group(P<0.05).Immunohistochemical positive results showed that red brown or brown staining of cytoplasm was positive expression.Conclusion Astragalus fermentation broth can significantly decrease the production of MDA and lipofuscin in the aging model of rats,and increase the level of GSH-Px,thus exerting an anti-aging effect,regardless of the dose.
