摘要
通过单因素试验和响应曲面法,以包封率和载药量为指标,研究槲皮素脂质体的最优处方。采用离心法分离槲皮素脂质体中游离的药物,高效液相色谱法测定药物含量,计算槲皮素脂质体的包封率;采用薄膜分散法制备槲皮素脂质体;以胆固醇和大豆磷脂的质量比、药脂比、水化温度为影响因素,采用响应曲面法进行处方筛选。结果表明,槲皮素脂质体优化后的处方为胆固醇和大豆磷脂的质量比1∶8,药脂比1∶24,水化温度50℃,在此条件下制备的槲皮素脂质体包封率为109.06%,载药量为4.54%。
The study on preparation technology of quercetin liposome was carried out by single factor and response surface methods with the entrapment efficiency and drug loading efficiency as indexes.The free drugs in quercetin liposome were separated by centrifugal method,and the drug content was determined by HPLC.Quercetin liposomes were prepared by membrane dispersion method.The ratio of cholesterol to soybean phospholipid,the ratio of liposome to soybean phospholipid and hydration or temperature were selected to perform the response surface method.The results showed that the optimized formulation of quercetin liposome were as follows:the mass ratio of cholesterol to soybean phospholipid 1∶8,the ratio of liposome to soybean phospholipid 1∶24,and the hydration temperature 50℃.Under these couditions,the eutrapment efficiency of quercetin liposome was 109.06%,and the drun loading was 4.54%.
