桑葚花色苷提取条件的响应面法优化 被引量:1

摘要 成熟桑葚中花色苷含量十分丰富,花色苷具有抗炎症、抗氧化、抗动脉粥样硬化、抗肿瘤等优良的生物学效应。将成熟桑葚冷冻干燥后对其花色苷含量进行分析,在对提取过程中的提取液酸化乙醇中乙醇浓度、乙醇与HCl体积比、提取温度、提取时间和提取次数进行单因素试验的基础上,采用响应面法对桑葚花色苷的提取条件进行了优化。结果表明:响应面法优化的桑葚花色苷提取条件为提取液酸化乙醇中乙醇浓度85%、乙醇与HCl体积比90∶10、提取温度60℃、提取时间90 min、提取1次,桑葚花色苷含量为2.22 mg/g。同时对不同种植条件下桑葚中的花色苷含量进行了差异性分析,为桑葚花色苷进一步开发利用提供了依据。 Mature mulberry is rich in anthocyanins that has the biological effects of anti-inflammatory,antioxidant,antiatherosclerosis and antitumor.The anthocyanins in freeze-dried mature mulberry was extracted,the ethanol concentration in the extract,the volume ratio of ethanol to HCl,extraction temperature,extraction time and extraction frequency were studied by single factor test,and the extraction conditions were optimized by the response surface method.The results showed that the ethanol concentration in the extract was determined to be 85%,the ratio of ethanol to HCl was 90∶10,the extraction temperature was 60℃,the extraction time was 90 min,the extraction frequency was once,and the anthocyanin content was 2.22 mg/g.The anthocyanin content in mulberry from different growing areas were analyzed,which provided a scientific basis for the further development and utilization of mulberry anthocyanins.
