摘要
本文评价了不同产地的山茶花提取液中活性物的含量和抗氧化能力。主要以不同产地的山茶花干花瓣为试验材料,以浓度75%的乙醇为溶剂,料液比为1︰15(w/v),采用超声工艺在温度为45℃下超声提取60 min后离心过滤浓缩,用紫外分光光度计等仪器测定了其主要活性成分—总黄酮、总多酚、总多糖和维生素C的含量。同时采用DPPH分析法和羟基自由基清除能力法,对四个产地的山茶花提取液的抗氧化能力进行了测试。结果表明:(1)不同产地的山茶花提取液中活性物含量和抗氧化能力存在显著差异,浙江大洋山的山茶花提取液中的总多酚、总黄酮、总多糖含量在这四个产地中都居于首位,维生素C含量和其它产地差异不大。(2)活性物中总多酚和总黄酮的含量与抗氧化能力存在明显的相关性,这两种活性物含量越高,抗氧化能力越强。(3)两种抗氧化性测试方法:DPPH分析法和羟基自由基清除能力法有较大相关性(R=0.955),可以从中任选一种,用来评价山茶花提取液的抗氧化性。此研究为山茶花产地的选择和其在化妆品、食品领域的开发利用提供了重要的依据。
In this paper,we studied the active ingredient contents and anti-oxidation ability of Camellia japonica flowers which are planted in four different areas.Ultrasound-assisted extraction was applied to extract active ingredients from Camellia japonica flowers at 45℃,60 min with 75%(v/v)ethanol,the ratio of Camellia japonica flowers to ethanol is 1︰15(w/v).After Centrifugation,filtration and concentration,the main active ingredient contents--total flavonoids,total polyphenols,total polysaccharides,and vitamin C were determined by uv-spectrophotometer.At the same time,the antioxidant activity was analyzed and compared by the scavenging capacity of(·OH)and DPPH experiment.The results showed that:(1)There were significant differences in the content of active substances and antioxidant capacity of camellia from four different planting areas.The camellia japonica flowers of ZheJiang province had high total flavonoids,total polyphenols,total polysaccharides,while the contents of vitamin C were not significantly different from other planting areas.(2)The higher the content of active substances such as total flavonoids and total polyphenols,the stronger the antioxidant capacity.(3)Two antioxidant test methods:hydroxyl radical(·OH)scavenging ability method and DPPH radical scavenging method have a large correlation(R=0.955),one of which can be selected to evaluate the antioxidant of these samples.This study provides an important basis for the selection of camellia planting area and its development and utilization in cosmetic and food fields.
