何首乌饮片的颜色与蒽醌类和二苯乙烯苷含量的相关性研究

Correlation between color and content of anthraquinones and stilbene glucoside of Ploygoni Multiflori Radix pieces

目的考察何首乌饮片的颜色与二苯乙烯苷、大黄素-8-O-β-D-葡萄糖苷、大黄素甲醚-8-O-β-D-葡萄糖苷、大黄素及大黄素甲醚5种成分含量之间的相关性,建立一种快速评价何首乌饮片质量的方法。方法观察何首乌饮片的外观性状,并采用分光测色仪测定其粉末颜色;采用超高效液相色谱法测定二苯乙烯苷、大黄素-8-O-β-D-葡萄糖苷、大黄素甲醚-8-O-β-D-葡萄糖苷、大黄素及大黄素甲醚的含量;运用典型相关分析方法对何首乌5种成分含量与颜色测定值进行相关性分析。结果随着炮制时间的延长,何首乌颜色变深,总色值E(E ab)减小,二苯乙烯苷和结合蒽醌含量总体呈降低趋势,游离蒽醌含量呈升高趋势,其颜色测定值与5种成分含量具有显著相关性(P<0.01)。结论何首乌炮制过程中颜色的变化可量化表达,E的大小在一定程度上能够反映何首乌饮片中二苯乙烯苷等5种成分的含量,色差检测是一种方便、快速评价何首乌饮片质量的方法。

Objective To determine the correlation between color and content of anthraquinones and stilbene glucoside of Ploygoni Multiflori Radix(PMR)pieces,based on the correlation between the two,and provide reference for a scientific method to quickly evaluate the quality of PMR pieces.Methods The appearance of PMR pieces was observed with eyes,and the color of powder was measured by colorimeter.Ultra-high performance liquid chromatography method was used to determine the content of stilbene glucoside and 4 kinds of anthraquinones.Canonical correlation analysis was used to explore the correlation between the color and the content of 5 chemical constituents of PMR pieces.Results As the processing time prolonged,the color of PMR pieces became darker,and E(E ab)decreased,and the content of stilbene glucoside and two combined anthraquinones decreased,and the content of two combined anthraquinone increased.There was significant correlation between the color of PMR pieces and the content of 5 chemical constituents(P<0.01).Conclusion The color change of PMR pieces can be quantified in the processing,and E reflects the content change in compositions of PMR pieces to some extent.E is an index to quickly determine the quality of PMR pieces,providing a reference for a convenient method to control the quality of PMR pieces in the processing.
