长链非编码RNA AC244502.3-201对甲状腺乳头状癌细胞增殖和迁移的影响

目的分析长链非编码RNA(long non-coding RNA,lncRNA)AC244502.3-201在甲状腺乳头状癌(papillary thyroid cancer,PTC)中的表达并探讨其对PTC细胞增殖、侵袭迁移的影响以及可能的分子机制。方法利用TANRIC数据库分析及实时荧光定量PCR(qRT-PCR)检测AC244502.3-201在PTC中的表达情况。Coding Potential Calculator软件预测编码能力,qRT-PCR检测亚细胞定位。Smart Silencer敲低细胞内AC244502.3-201表达后,CCK-8法、EdU实验和克隆形成实验检测细胞增殖能力,Transwell实验检测侵袭迁移能力。qRT-PCR检测对NF-κB下游基因的影响,双荧光素酶报告实验和Western Blot实验检测NF-κB通路激活情况。结果在PTC中AC244502.3-201的表达显著升高。AC244502.3-201不具有编码蛋白的能力,主要定位于细胞浆(P<0.05)。敲低AC244502.3-201能抑制细胞增殖、侵袭和迁移(P<0.05)。敲低AC244502.3-201抑制了p65入核、NF-κB信号通路活性以及下游基因表达。结论在PTC中AC244502.3-201表达升高。敲低AC244502.3-201抑制了NF-κB通路激活及下游基因表达,最终抑制PTC细胞的增殖、侵袭迁移能力。

Objective To analyze the expression of long non-coding RNA(lncRNA)AC244502.3-201 in papillary thyroid cancer(PTC)and explore its effect on PTC cell proliferation,invasion and migration,and the underlying molecular mechanism.MethodsTANRIC database and real-time fluorescent quantitative PCR(qRT-PCR)were used to detect the expression of AC244502.3-201 in PTC.Coding Potential Calculator software predicts coding capabilities and qRT-PCR detects subcellular localization.After Smart Silencer knocked down the expression of AC244502.3-201 in cells,CCK-8 method,EdU test and clone formation test were used to detect cell proliferation,and Transwell test was used to detect invasion and migration.qRT-PCR was used to detect the effects of NF-κB downstream genes.The double luciferase reporter assay and Western Blot assay were used to detect NF-κB pathway activation.ResultsThe expression of AC244502.3-201 was significantly increased in PTC.Knockdown of AC244502.3-201 inhibited the cell proliferation,invasion and migration(P<0.05).Knockdown of AC244502.3-201 inhibited the nuclear translocation of p65,NF-κB transcriptional activity andthe expression of downstream genes(P<0.05).ConclusionsAC244502.3-201 was highly expressed in PTC.Knockdown of AC244502.3-201 suppressed the proliferation and migration of PTC cells through the inhibition of NF-κB signaling.
