摘要
目的探讨miR-200c在子宫内膜腺癌血清及组织中的表达及临床意义。方法采用RT-PCR方法检测120例子宫内膜腺癌患者和100例对照组增生期或萎缩型子宫内膜患者血清和组织中的miR-200c表达水平,分析血清和组织中的miR-200c与子宫内膜腺癌临床病理特征的关系,应用ROC曲线评价组织和血清中miR-200c对子宫内膜腺癌诊断的灵敏度和特异度。结果子宫内膜腺癌组织和血清中miR-200c相对表达量显著高于对照组(P <0.05)。miR-200c在临床晚期、深肌层浸润和存在淋巴结转移的子宫内膜腺癌组织中表达明显下降(P <0.05),而在临床晚期、深肌层浸润和存在淋巴结转移的子宫内膜腺癌患者血清中表达增高(P <0.05)。miR-200c血清和组织中诊断子宫内膜腺癌的敏感度分别为:76.5%和77.1%;特异性分别为44.7%和30.73%。结论 miR-200c作为诊断子宫内膜腺癌的分子标志物敏感性较高,但特异度低。
ObjectiveTo investigate the expression and clinical significance of miR-200 c in serum and tissues of endometrial adenocarcinoma.MethodsTo use RT-PCR method to detect the expression of miR-200 c in tissues and serum of 120 patients with endometrial adenocarcinoma and 100 cases of control group,analyze the relationship between the expression of miR-200 c and the features of clinic pathology in endometrial adenocarcinoma and apply ROC curve to evaluate the sensitivity and specificity of mir-200 c in the diagnosis of endometrial adenocarcinoma.Results The relative expression of miR-200 c in tissues and serum of endometrial adenocarcinoma was significantly higher than those in the control group(P < 0.05). The expression of miR-200 c was significantly decreased in endometrial carcinoma tissues with advanced clinical stage,deep muscular layer infiltration and lymphnode metastasis(P < 0.05). However,in patients with advanced clinical stage,deep muscular layer infiltrationand endometrial carcinoma with lymph node metastasis,serum expression were increased(P < 0.05). The sensitivity of miR-200 c in serum and tissue for diagnosis of endometrial adenocarcinoma was 76.5% and 77.1% respectively,and the specificity was 44.7% and 30.73% respectively. Conclusion:As a molecular marker for the diagnosis of endometrial adenocarcinoma,the sensitivity of miR-200 c is high,however,the specificity is low.
