益胃饮中药血清药物化学研究 被引量:4

摘要 目的对临床经验方益胃饮开展中药血清药物化学研究,阐明其体内药效物质基础,为益胃饮的药物开发和临床应用提供科学依据。方法采用超高效液相色谱串联四级杆飞行时间质谱(UHPLC-Q/TOF-MS)快速鉴定益胃饮水提物化学成分,及其经大鼠灌胃给药后吸收入血的移行成分。采用ACQUITY UPLC HSS T3(100×2.1 mm,1.8μm)色谱柱,以0.1%甲酸乙腈-0.1%甲酸水为流动相进行梯度洗脱,采用电喷雾电离技术,正负离子模式扫描,对益胃饮水提物、空白血清及大鼠灌胃益胃饮水提物后的含药血清进行对比分析,确定血中移行成分。结果通过SCIEX OS软件比较色谱峰保留时间、化合物精确分子量以及二级质谱裂解规律等方式,共归属益胃饮化学成分88个,益胃饮含药血清移行成分27个。结论建立的基于UHPLC-Q/TOF-MS的益胃饮中药血清药物化学分析方法简便、快速、准确,为揭示益胃饮防治胃癌前病变及胃癌的药效物质基础提供实验依据。 Objective To carry out the research on traditional Chinese medicine serum pharmacochemistry for clinical prescription Yiwei Decoction,and to clarify the pharmacodynamic substance basis of Yiwei Decoction,and to provide scientific basis for drug development and clinical application of Yiwei Decoction.Methods UHPLC-Q/TOF-MS was used to rapidly identify the chemical constituents of Yiwei Decoction water extract and its constituents absorbed into the blood after intragastric administration in rat.Utilizing ACQUITY UPLC HSS T3(100×2.1 mm,1.8μm)column,gradient elution with 0.1%formic acid acetonitrile-0.1%formic acid as mobile phase and electrospray ionization technique with positive and negative ion mode scanning,the Yiwei Decoction water extract,blank serum and drug serum were detected and analyzed to identify the blood migration components.Results The SCIEX OS software was used to compare the retention time,precise molecular weight and mass spectrometry fragmentation to character 88 chemical constituents and 27 constituents absorbed into the blood of Yiwei Decoction.Conclusion The established traditional Chinese medicine serum pharmacochemistry method based on UHPLC-Q/TOF-MS is simple,rapid and accurate,which could provide the experimental basis for revealing the pharmacodynamic basis of Yiwei Decoction for preventing and treating precancerous lesions and gastric cancer.
