摘要
目的:探讨熊果酸(UA)对胶原诱导性关节炎(CIA)大鼠关节软组织RANKL、RANK、OPG mRNA表达的影响。方法:选取雌性SD大鼠复制CIA模型,造模成功后随机分为CIA模型组、熊果酸低剂量组、熊果酸高剂量组、甲氨蝶呤组、熊果酸+甲氨蝶呤组,并给予相应处理。于初次免疫30d后处死大鼠,取炎症关节进行研究,荧光定量PCR法检测RANKL、RANK、OPG在大鼠关节匀浆中的表达。结果:与CIA模型组比较,UA大鼠高剂量组关节RANKL mRNA的表达明显降低(P<0.01);与MTA组比较,UA+MTA组关节RANKL mRNA的表达明显降低(P<0.05)。结论:UA影响CIA大鼠关节软组织RANKL mRNA的表达,其机理可能与抗炎作用有关。
Objective:To investigate the effects of ursolic acid(UA)on the expression of RANKL,RANK,OPG mRNA in the soft tissues of collagen-induced arthritis(CIA)rats.Methods:Female SD rats were selected to reproduce the CIA model.After successful modeling,they were randomly divided into the CIA model group,ursolic acid low-dose group,ursolic acid high-dose group,methotrexate group,ursolic acid+methotrexate Group and give appropriate treatment.Rats were sacrificed 30 days after the first immunization,and inflammatory joints were taken for research.The expressions of RANKL,RANK,OPG,and rat joint homogenate were detected by fluorescent quantitative PCR.Results:Compared with the CIA model group,the expression of RANKL mRNA in joints of UA rats in the high-dose group was significantly reduced(P<0.01);compared with the MTA group,the expression of RANKL mRNA in joints in the UA+MTA group was significantly reduced(P<0.05).Conclusion:UA affects the expression of RANKL mRNA in articular soft tissues of CIA rats,and its mechanism may be related to anti-inflammatory effects.
