摘要
为开发低温厌氧消化过程中产甲烷微菌资源,该试验采用亨盖特厌氧分离技术和16S rDNA测序技术,从延边地区低温期的湖底底泥中筛选出1株产甲烷微菌BDP-8,该菌株最适合的生长温度为15℃,pH值7.0,NaCl浓度为0~0.4 mol/L,该菌株只能利用H2/CO2(80∶20)混合气体。16S rDNA的系统发育分析鉴定为产甲烷微菌的Methanolaciniapetrotearia菌株。
In order to exploit the methanomicrobia resource in low temperature physchrophilic anaerobic digestion process,a methanogen BDP-8 was isolated from different lake sludge of Yanbian area in low temperature phase by Hengate anaerobic separation technology and 16S rDNA gene sequencing technology.The optimal growth conditions of BDP-8 were temperature 15℃,pH 7.0,the concentration of NaCl 0~0.4 mol/L.The strain only used hydrogen and carbon dioxide(80:20).It was identified as methanolaciniapetrotearia strain by 16S rDNA gene phylogenetic analysis.
