摘要 为对鹿筋胶原蛋白提取条件进行优化,该试验采用单因素分析及正交试验筛选出鹿筋胶原蛋白提取条件,以及提取产物对体外培养MC3T3成骨细胞的增殖作用。结果表明:最佳提取条件为醋酸体积分数5%,浸泡时间48 h,煎煮时间1.5 h,胃蛋白酶用量1∶1000,酶解时间6 h,鹿筋胶原蛋白回收率58.56%,蛋白质含量48.4%。优化后的条件下提取的鹿筋胶原蛋白对MC3T3细胞具有显著的增殖作用,其增殖作用呈量效关系,当浓度为8 mg/mL时,增殖作用最为显著。因此,鹿筋胶原蛋白提取条件优化后,回收率增加;鹿筋胶原蛋白对体外培养的成骨细胞具有明显的增殖作用,为其产品的开发提供理论依据。 In order to optimize the extraction conditions of deer sinew collagen,single factor analysis and orthogonal experiments were used to screen out the conditions of deer sinew collagen extraction and the effect of the extracted products on MC3T3 osteoblasts in vitro.The results showed that the optimal extraction conditions were 5% acetic acid volume fraction,soaking time was 48 h,decoction time was 1.5 h,pepsin dosage was 1∶1000,enzymolysis time was 6 h,and deer tendon collagen recovery was 58.56%,the protein content was 48.4%.The deer tendon collagen extracted under optimized conditions had a significant proliferative effect on MC3T3 cells,and its proliferative effect was in a dose-effect relationship.When the concentration was 8 mg/mL,the proliferative effect was the most significant.Conclusion:the optimized extraction conditions of deer sinew collagen increase the recovery rate;deer sinew collagen has a significant proliferative effect on osteoblasts cultured in vitro,providing a theoretical basis for the development of its products.
