摘要
昼夜节律可维持机体的多种生理功能,调节机体应对外界环境变化。昼夜节律紊乱则会产生不良影响并增加患病风险。越来越多的研究表明,很多小分子调节剂可靶向作用于昼夜节律系统,高通量筛选等新技术新策略也已用于发现和研究更多新的小分子昼夜节律调节剂。本综述介绍了昼夜节律系统的基本组成,运行的分子机制以及生物钟小分子调节剂的研究进展,并阐述了各种慢性疾病与生物钟的关系,以及生物钟小分子调节剂对这些疾病的治疗潜力。
The circadian rhythm maintains various physiological functions of our body,and regulates the body to cope with the external environment. The disorder in the circadian rhythm could produce harmful effects and increase the risk of diseases. A growing research has shown that many small molecule regulators could target the circadian rhythm system,and many new techniques and strategies,including the high-throughput screening technique,have been used in the discovery and investigation of new small molecule regulators of the circadian rhythm system. In this review,we introduce basic components and molecular mechanisms of the circadian rhythm system,summarize research progresses in the small molecule regulators,explain the relationship between various chronic diseases and circadian clocks,and discuss potential applications of the small molecule modulators in the treatment of circadian rhythm-related diseases.
