摘要
目的探讨多囊卵巢综合征(PCOS)患者血清特异性长链非编码核糖核酸(LncRNA)表达谱及下游mRNA通路。方法建立PCOS患者(PCOS组,50例)和健康人群(对照组,50例)队列,随机选取各3例,采用单色微阵列基因表达分析流程进行表达谱检测,并辅以热图分析、实时定量逆转录-聚合酶链反应(qRT-PCR)验证和下游mRNA的基因本体注释(GO)富集分析。结果PCOS患者血清中分别有351、217种显著上调和下调的LncRNA(符合变化系数≥2.0,P<0.05);进一步聚类分析发现,包含显著上调的11种LncRNA和显著下调的8种LncRNA的数据集可以成功区分PCOS患者和健康人群。qRT-PCR成功验证其中16种LncRNA具有相同显著变化。GO富集分析发现,这些LncRNA调控的mRNA功能范围包括细胞活化/生长/周期、基因及表观遗传学负调控等方面。结论 PCOS患者血清特异性LncRNA表达谱可为疾病的非侵入性诊断提供分子生物学标志物。
Objective To investigate the expression profile of serum LncRNA and the downstream mRNA pathway in patients with polycystic ovary syndrome(PCOS). Methods Fifty patients with PCOS and 50 healthy controls were enrolled in the study. The expression profile of LncRNA from randomly selected 3 PCOS patients and 3 healthy controls was detected by single color microarray gene expression analysis, supplemented by thermogram analysis, qRT-PCR verification and gene ontology(GO) analysis of downstream mRNA. Results Total 351 up-regulated and 217 down-regulated serum LncRNA were detected in PCOS patients, comparing with healthy controls(the coefficient of change was≥2.0, P<0.05). Further cluster analysis showed that the data set containing 11 up-regulated and 8 down-regulated LncRNA could successfully distinguish PCOS from normal control, where 16 similar tendency were further verified by qRT-PCR. GO analysis showed that the mRNA functions regulated by these LncRNA included cell activation/growth/cycle, gene and epigenetic negative regulation and so on. Conclusion The expression profile of serum LncRNA has the potential to provide molecular biomarkers for noninvasive diagnosis of PCOS.
