摘要
基因工程学实验课是生物技术专业的主要实践课程之一,为培养大学生的创新意识和科研能力,以新孢子虫NcAMA1基因的扩增、克隆及真核表达为例,开展基因工程学实验教学设计。实践表明,通过对基因组DNA提取、目的基因扩增、克隆载体与表达载体构建、以及目的基因表达等实验步骤的实践操作,激发了学生的科研兴趣和创造力,培养了学生分析问题和解决问题的能力,提升了学生的实验技能、创新意识和综合素质。
The course of gene engineering experiment is one of the main practice courses of biotechnology major. In order to cultivate the innovative consciousness and scientific research ability of college students,in this study,the amplification,cloning and eukaryotic expression of Nc AMA1 gene in Neospora caninum were taken as examples to carry out the experimental teaching design of genetic engineering. Practice shows that through practicing the experimental steps of genomic DNA extraction,target gene amplification,cloning vector and expression vector construction,and target gene expression,students’ interest in scientific research and creativity is stimulated. Their ability of analyzing and solving problems is cultivated. At the same time,their experimental skills,innovation awareness and comprehensive quality are improved.
