从前额皮质星形胶质细胞、IgG及IL-2探讨孕期恐应激对其近远期仔鼠情绪的影响

Effects of prefrontal astrocyte and IgG and IL-2 in fear-impaired pregnant rats the emotions of young and adult offspring rats

目的:从前额皮质星形胶质细胞和免疫球蛋白G(IgG)、白细胞介素-2(IL-2)探讨孕期恐应激对其近远期仔鼠情绪的影响。方法:运用旁观电击法制备恐伤孕鼠模型,通过旷场实验检测其21、80日龄仔鼠情志变化,采用免疫组化法检测前额叶皮质胶质纤维酸性蛋白(GFAP)表达,ELISA法检测血液中IgG、IL-2的含量。结果:与同日龄对照组比较,模型组21、80日龄仔鼠垂直得分、水平得分,糖水消耗量、糖水偏好值均显著降低(P<0.05);与同日龄对照组比较,模型组21、80日龄仔鼠血清IgG、IL-2含量均显著降低,前额叶皮质GFAP蛋白表达均上调(P<0.05)。结论:孕期恐应激可导致其近远期仔鼠情志异常,可能与星形胶质细胞过度激活和免疫异常有关。

Objective:To explore the effects of fear-impaired pregnant rats on the emotions of young and adult offspring rats from prefrontal astrocyte,IgG and IL-2.Methods:The rat model of pregnancy with fear injury was prepared by the bystander electric shock method,emotional changes were observed by open field test and sucrose preference test on the 21-day-old and 80-day-old neonatal rats,the GFAP expression in PFC was assayed by immunohistochemistry,the contents of IgG and IL-2 in blood were detected by ELISA.Results:Compared with the control group,the model group at 21 and 80 days old pups had lower vertical scores and horizontal scores,less sugar and water consumption,and lower sugar and water preference values(P<0.05).Compared with the control group,IgG and IL-2 of the 21-day-old and 80-day-old offspring rats in the model group were decreased,and the expression of GFAP protein in the prefrontal cortex was up-regulated,the difference was statistically significant(P<0.05).Conclusion:The fear-impaired pregnant rats could cause emotional changes in young and adult offspring rats,which may be associated with excessive activation of astrocytes and immune abnormalities.
