益气解毒方通过PI3K/AKT/mTOR信号通路诱导鼻咽癌细胞自噬的研究 被引量:6

Study on Yiqi Jiedu Formula inducing autophagy in nasopharyngeal carcinoma cells through PI3K/AKT/mTOR signaling pathway

摘要 目的:探讨益气解毒方对鼻咽癌细胞自噬的影响及其机制。方法:MT T检测益气解毒方对鼻咽癌CNE-2Z细胞增殖的影响。透射电镜和MDC染色观察益气解毒方诱导CNE-2Z细胞的自噬效应。Western Blot法检测Beclin-1、LC3、PI3K、p-AKT和p-mTOR蛋白表达量。结果:益气解毒方抑制CNE-2Z细胞增殖(P<0.05,P<0.01)。与对照组比较,不同浓度益气解毒方组细胞内出现自噬体、MDC荧光强度增加、Beclin-1和LC3Ⅱ蛋白表达上调(P<0.05,P<0.01),其中0.5mg/mL益气解毒方组增加最明显。与0.5mg/mL益气解毒方组比较,0.5mg/mL益气解毒方+3-MA组MDC荧光强度、Beclin-1和LC3Ⅱ蛋白降低(P<0.05)。与对照组比较,0.5mg/mL和1mg/mL益气解毒方组的PI3K、p-AKT和p-mTOR的蛋白表达均出现下调(P<0.05)。结论:益气解毒方可诱导鼻咽癌CNE-2Z细胞自噬,其机制可能是通过激活PI3K/AKT/mTOR信号通路。 Objective:To investigate the effects of Yiqi Jiedu Formula on autophagy of nasopharyngeal carcinoma cells and its mechanism.Methods:MTT was used to detect the effects of Yiqi Jiedu Formula on the proliferation of nasopharyngeal carcinoma CNE-2Z cells.Transmission electron microscopy and MDC staining were used to observe the autophagic effect of Yiqi Jiedu Formula on CNE-2Z cells.Western Blot was used to detect the expression levels of Beclin-1,LC3,PI3K,p-AKT and p-mTOR proteins in CNE-2Z cells treated with Yiqi Jiedu Formula.Results:MTT results showed that Yiqi Jiedu Formula had a significant inhibitory effect on the proliferation of CNE-2Z cells(P<0.05,P<0.01).Compared with the control group,autophagosome,MDC fluorescence intensity increased,Beclin-1 and LC3Ⅱprotein expression increased in Yiqi Jiedu Formula group(P<0.05,P<0.01),and autophagosome,fluorescence intensity,Beclin-1 and LC3Ⅱprotein content increased significantly in 0.5mg/mL Yiqi Jiedu Formula group.Compared with 0.5mg/mL Yiqi Jiedu Formula group,the fluorescence intensity of MDC,Beclin-1 and LC3II protein in 0.5 mg/mL Yiai Jiedu Formula+3-MA group decreased(P<0.05).Compared with the control group,the expression of PI3K,p-AKT and p-mTOR in 0.5mg/mL and 1mg/mL Yiqi Jiedu Formula groups decreased(P<0.05).Conclusion:Yiqi Jiedu Formula can induce autophagy in CNE-2Z cells of nasopharyngeal carcinoma through activating PI3K/AKT/mTOR signaling pathway.
