摘要
目的:观察广嗣育麟汤对雷公藤多苷(GTW)诱导的生精障碍大鼠Bcl-2、Bax、c-kit、Oct4蛋白及mRNA表达的影响。方法:60只SD大鼠随机分为空白对照组、模型对照组和中药干预组,GTW灌胃建立大鼠生精障碍模型,中药干预组予广嗣育麟汤灌胃,4周后用免疫组化法检测大鼠睾丸组织Bcl-2、Bax,Western Blot以及Real-time PCR检测睾丸组织c-kit、Oct4蛋白及mRNA的表达。结果:与模型对照组比较,中药干预组Bax蛋白含量显著降低、Bcl-2的蛋白含量显著升高(P<0.01),c-kit蛋白和mRNA表达量显著升高(P<0.05),Oct4的蛋白和mRNA表达量也有提升,但差异无统计学意义。结论:广嗣育麟汤可以有效调控模型大鼠Bax、Bcl-2、c-kit蛋白及其mRNA的表达,具有抑制生精细胞凋亡、促进生精细胞增殖作用。
Objective:To observe the effects of Guangsi Yulin Decoction on expression of Bcl-2,Bax,c-kit and Oct4 protein and mRNA in rats with Tripterygium wilfordii(GTW)-induced spermatogenic disorder.Methods:Sixty rats were randomly divided into control group,GTW group and Guangsi Yulin group.The spermatogenic disorder rats were established by GTW.All administration lasted for 4 weeks.The expression of Bax,Bcl-2,c-kit and Oct4 in rat testis was detected by immunohistochemistry,Western Blot and Real-Time PCR.Results:Compared with model group,the protein expression of Bax was decreased and Bcl-2 was increased in Guangsi Yulin group(P<0.01).Western Blot and Real-time PCR results showed that the protein and mRNA expression of c-kit in Guangsi Yulin group was higher than that in GTW group(P<0.05).The protein and mRNA expression of Oct4 also increased in Guangsi Yulin group compared with GTW group,although with no significant differences.Conclusion:Guangsi Yulin Decoction can effectively regulate the protein and mRNA expression of Bax,Bcl-2 and c-kit,and has the function of inhibiting apoptosis of spermatogenic cell and promoting its proliferation.
