摘要
目的:探讨肾衰Ⅱ号方对5/6(A/I)肾切除慢性肾功能衰竭(CRF)模型大鼠贫血的影响。方法:32只SD大鼠制备5/6(A/I)肾切除CRF大鼠模型,取全血及残余肾组织标本,测定血清肌酐(Scr)、尿素氮(BUN),Masson染色观察肾组织病变,血常规,残余肾组织缺氧诱导因子-2α(HIF-2α)、促红细胞生成素(EPO)及铁调素25(Hepcidin-25)蛋白表达。结果:与模型组比较,肾衰Ⅱ号方显著降低大鼠Scr、BUN水平(P<0.05),减轻肾间质炎性细胞浸润及胶原纤维增生,显著升高外周血RBC、Hb、HCT水平(P<0.05),显著上调残肾组织中HIF-2α、EPO,同时下调Hepcidin-25蛋白表达(P<0.05)。结论:具有益气养血、化瘀泄浊作用的肾衰Ⅱ号方通过保护肾固有细胞,增加内源性EPO的合成,调节铁代谢紊乱,减轻肾性贫血,改善肾间质纤维化。
Objective:To investigate the effect of ShenshuaiⅡDecoction on anemia in rats induced by 5/6th(ablation/infarction,A/I)nephrectomy.Methods:Thirty-two SD rats were used to prepare the chronic renal failure(CRF)rat model by means of 5/6(A/I)nephrectomy.Take the blood and remnant nephridial tissue to investigate:serum creatinine(Scr),blood urea nitrogen(BUN);Masson staining to observe renal tissue lesions;blood routine;protein expressions of HIF-2α,EPO,Hepcidin-25 in remnant nephridial tissue.Results:Compared with model group,the levels of Scr and BUN were significantly decreased(P<0.05),and the inflammatory cell infiltration and collagen fibrosis in the renal interstitial were alleviated in ShenshuaiⅡDecoction group.Moreover,the RBC,Hb,HCT levels were significantly increased(P<0.05),and the protein expressions of HIF-2α,EPO in remnant nephridial tissue of ShenshuaiⅡDcoction group rats were increased while the expression of Hepcidin-25 was decreased(P<0.05).Conclusion:ShenshuaiⅡDecoction could increase the synthesis of endogenous EPO and regulate iron metabolism disorder,thus alleviated renal anemia and interstitial fibrosis.
