清润养目口服液对肝肾阴虚模型兔血清激素水平及睫状肌组织中cAMP、cGMP含量的影响研究 被引量:8

摘要 目的:通过对肝肾阴虚模型兔血清激素水平及睫状肌组织中环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)含量的检测,探讨清润养目口服液缓解症状性视疲劳的可能机制。方法:选取健康日本大耳白兔30只,随机分为正常组、模型组、杞菊地黄组和清润养目高、低剂量组5组,每组6只。除正常组外,其余各组均以15 mg/kg的剂量经口灌胃甲状腺素,然后以0.100 mg/kg的剂量肌注利血平制备肝肾阴虚模型,采用白炽灯昼夜光照循环,昼30照明度(lux),夜0 lux,连续照射7 d后分别给药治疗2周。杞菊地黄组按2.500 mL/kg的剂量给药,清润养目高、低剂量组分别按2.300、0.575 mL/kg的剂量给药。在末次给药后的24 h,麻醉、处死实验兔,眼眶后静脉丛取血后迅速摘出双眼眼球,剥离睫状体,制作标本。用ELISA法测定兔血清雌二醇(E2)、睾酮(T)、皮质酮(CORT)及眼睫状肌中cAMP、cGMP含量。结果:与正常组比较,模型组兔血清T、CORT、眼睫状肌组织中cAMP含量显著降低,E2、E2/T、cGMP含量显著升高,差异均有统计学意义(P<0.05)。与模型组比较,清润养目高剂量组、杞菊地黄组血清T均升高(P<0.05);E2、E2/T均降低(P<0.05);清润养目低剂量组差异无统计意义;杞菊地黄组、清润养目高、低剂量组血清CORT、cAMP均升高(P<0.05),cGMP降低(P<0.05)。与杞菊地黄组比较,清润养目高剂量组血清E2、T、E2/T、CORT差异均无统计学意义,cAMP较高(P<0.05),cGMP较低(P<0.05);清润养目低剂量组血清E2、E2/T含量均较高(P<0.05),血清T、CORT,cAMP含量较低(P<0.05),cGMP较高(P<0.05)。结论:清润养目口服液能有效恢复肝肾阴虚模型兔血清CORT水平和降低E2水平,升高T水平,缩小E2/T值及升高睫状肌组织中cAMP含量,降低cGMP含量,从而有效缓解肝肾阴虚表现及视疲劳症状。 Objective: To discuss the possible mechanism of Qingrun Yangmu oral liquid in reliving symptomatic asthenopia by detecting the levels of hormones in serum and the contents of cyclic adenosine monophosphate(cAMP) and cyclic guanosine monophosphate(cGMP) in ciliary tissue in model rabbits with liver-kidney yin deficiency. Methods:A total of30 healthy Japanese white rabbits were selected and randomly divided into the normal group, the model group, the Qiju Dihuang group and the Qingrun Yangmu groups of high-dose and low-dose,six rabbits in each group. Except in the normal group,models of liver-kidney deficiency were established in other groups by giving 15 mg/kg of thyroxine by gavage through mouth,then 0.100 mg/kg of reserpine by intramuscular injection;these models were exposed to a light-dark cycle with incandescent lamp, 30 lux in daytime and 0 lux at night, and after seven days of continuous exposure, they were given administration respectively for two weeks. The dosage of administration was 2.500 mL/kg in the Qiju Dihuang group,2.300 mL/kg in the Qingrun Yangmu group of high-dose and 0.575 mL/kg in the Qingrun Yangmu group of low-dose. The experimental rabbits were anesthetized and killed 24 hours after the last administration. After blood collection from the orbital venous plexus, the eyeballs were quickly removed and the ciliary bodies were detached to make specimens. The ELISA method was applied to detect the contents of estradiol(E2),testosterone(T) and corticosterone(CORT) in serum,as well as cAMP and cGMP in ciliary tissue. Results:When compared with those in the normal group,the contents of T and CORT in serum as well as cAMP in ciliary tissue in the model group were significantly decreased,and the contents of E2,E2/T and cGMP were significantly increased,differences being significant(P<0.05). When compared with that in the model group,the contents of T were increased in both the Qingrun Yangmu group of high-dose and the Qiju Dihuang group(P<0.05),and the contents of E2 and E2/T were decreased(P<0.05);there was no significant difference being found in the comparison between the model group and the the Qingrun Yangmu group of low-dose;the contents of CORT in serum and cAMP in the Qiju Dihuang group and the Qingrun Yangmu groups of high-dose and low-dose were increased(P<0.05), and cGMP were decreased(P<0.05). Compared the contents of E2,T,E2/T and CORT in serum,there was no significant difference being found between the Qiju Dihuang group and the Qingrun Yangmu group of high-dose;the content of cAMP in the Qingrun Yangmu group of high-dose was higher(P<0.05),and the content of cGMP was lower(P<0.05). When compared with those in the Qiju Dihuang group,the contents of both E2 and E2/T in serum in the Qingrun Yangmu group of low-dose were higher(P<0.05), and the contents of T, CORT and cAMP were lower(P<0.05), the content of cGMP was higher(P<0.05).Conclusion:For model rabbits with liver-kidney yin deficiency,Qingrun Yangmu oral liquid can effectively restore the level of CORT,reduce the level of E2,increase the level of T and reduce the E2/T value in serum,as well as increase the content of cAMP and reduce the content of cGMP in ciliary tissue, thereby it can effectively relieve the liver-kidney deficiency and asthenopia.
