摘要
目的探究高脂血症颈动脉粥样硬化家兔NLRP3炎性小体及相关炎症因子的表达水平。方法将20只新西兰大白兔随机分为对照组和模型组,每组10只,适应性喂养一周后模型组行颈动脉空气干燥术,并予高脂饲料饲喂养,对照组予普通饲料喂养。4周后处死2组家兔,取血清及颈动脉组织进行相关检测并进行统计分析。结果模型组血清胆固醇、三酰甘油较正常组显著上升;模型组颈动脉组织蛋白p-P65核因子-κB(NF-κB)、NLRP3、天冬氨酸蛋白水解酶-1(Caspase-1)、白介素-1β(IL-1β)及白介素-18(IL-18)较正常值组表达显著上升,差异有统计学意义(P均<0.001);模型组血清蛋白NF-κB、IL-1β及IL-18的表达较正常值组表达显著上升,差异有统计学意义(P均<0.001)。结论高脂血症颈动脉粥样硬化家兔模型存在NLRP3及进相关炎症因子如caspase-1、IL-1β和IL-18的高表达,且活化NLRP3炎症小体促进动脉粥样硬化(AS)机制可能与NF-κB磷酸化有关。
Objective To investigate the expression of NLRP3 inflammasome and related inflammatory factors in hyperlipidemia carotid atherosclerosis rabbits.Methods 20 New Zealand white rabbits were randomly divided into control group and model group,with 10 rabbits in each group.After one week of adaptive feeding,the model group was given carotid air drying and fed with high-fat diet,while the control group was fed with ordinary diet.After 4 weeks,2 groups of rabbits were killed,serum and carotid artery tissues were collected and analyzed statistically.Results Serum cholesterol and triglyceride in the model group were significantly higher than those in the normal group.The protein expressions of carotid artery tissue p-P65 NF-κB,NLRP3,Caspase-1,IL-1βand IL-18 in the model group were significantly increased compared with the normal group,with statistically significant differences(P<0.001).Moreover,the expressions of serum proteins NF-κB,IL-1βand IL-18 in the model group were significantly increased compared with the normal group,with statistically significant differences(P<0.001).Conclusion There are high expression of NLRP3 and related inflammatory factors such as caspase-1,IL-1βand IL-18 in hyperlipidemia carotid artery atherosclerosis in rabbits,and the AS mechanism promoted by activating NLRP3 inflammasome may be related to the phosphorylation of NF-κB.
