健脾理气颗粒对脾虚气滞证功能性消化不良大鼠胃肠运动功能及胃肠激素的影响 被引量:11

摘要 目的分析健脾理气颗粒对脾虚气滞证功能性消化不良(FD)大鼠胃肠运动功能及胃肠激素的影响。方法将73只SD大鼠随机分为空白组(13只)和健脾理气颗粒低、中、高剂量组(各12只)、西药组(12只)、模型组(12只)。除空白组外,其他组均采用碘乙酰胺+站小平台法+隔日禁食建立脾虚气滞证FD大鼠模型。健脾理气颗粒各剂量组分别给予相应剂量健脾理气颗粒灌胃,西药组给予莫沙必利灌胃,空白组和模型组灌胃等量生理盐水。比较6组大鼠造模前后、治疗前后体重变化情况,治疗后的胃排空率、血清胃肠激素及炎症因子水平。结果造模后,其他5组大鼠的体重增加均少于空白组大鼠(均P<0.05);药物治疗后,健脾理气颗粒中、高剂量组和西药组大鼠的体重增加均大于模型组(均P<0.05)。模型组的胃排空率低于空白组,而健脾理气颗粒各剂量组和西药组大鼠的胃排空率均高于模型组(P<0.05)。与空白组比较,模型组大鼠血清中胃促生长素(Ghrelin)、胃动素水平降低,血管活性肠肽(VIP)、胆囊收缩素八肽(CCK8)水平升高(均P<0.05);与模型组比较,西药组和健脾理气颗粒中、高剂量组血清VIP、CCK8水平降低而Ghrelin、胃动素水平升高,健脾理气颗粒低剂量组血清CCK8水平亦降低(均P<0.05)。6组的白细胞介素6和白细胞介素10水平差异均无统计学意义(均P>0.05)。结论健脾理气颗粒均能促进脾虚气滞证FD大鼠胃排空及体重增长,纠正胃肠激素紊乱;其或通过调节脑-肠的动态平衡而有效改善胃肠动力。 Objective To analyze the effect of Jianpi Liqi Granules on gastrointestinal motility and gastrointestinal hormones in rats suffering from functional dyspepsia(FD) with spleen deficiency and qi stagnation syndrome. Methods Seventy-three SD rats were randomly divided into blank group(n=13), low-dose Jianpi Liqi Granules group(n=12), medium-dose Jianpi Liqi Granules group(n=12), high-dose Jianpi Liqi Granules group(n=12), western medicine group(n=12) and model group(n=12). Except the blank group, the other groups adopted iodoacetamide + small platform method + every other day fasting to establish FD rat model with spleen deficiency and qi stagnation syndrome. The Jianpi Liqi Granules groups received intragastric administration of Jianpi Liqi Granules with corresponding dose, the western medicine group received intragastric administration of mosapride, and the blank and model groups received intragastric administration of equivalent volume of normal saline. Changes in weight before and after modeling or before and after treatment, and post-therapeutic gastric emptying rate, serum gastrointestinal hormones and inflammatory factors levels were compared among the six groups. Results After modeling,the weight gain of the rats were less in the remaining five groups than in the blank group(all P<0.05);after medication, the weight gain of the rats were increased in the medium-and high-dose Jianpi Liqi Granules groups and the western medicine group than in the model group(all P<0.05). The model group yielded a lower gastric emptying rate than the blank group, whereas each Jianpi Liqi Granules group and the western medicine group obtained a higher gastric emptying rate than the model group(P<0.05). Compared with the blank group, the model group possessed lower Ghrelin and motilin levels and higher vasoactive peptide(VIP) and cholecystokinin 8(CCK8) levels(all P<0.05);compared with the model group, the western medicine group and the medium-and high-dose Jianpi Liqi Granules groups exhibited reduced serum VIP and CCK 8 levels and elevated Ghrelin and motilin levels, and the low-dose Jianpi Liqi Granules group also obtained a lower serum CCK8 level(all P<0.05). There were no statistically significant differences in interleukin 6 or interleukin 10 level among the six groups(all P>0.05). Conclusion Jianpi Liqi Granules can promote gastric emptying and weight gain and correct gastrointestinal hormones disorder in FD rats with spleen deficiency and qi stagnation syndrome;it may improve gastrointestinal motility effectively by regulating brain-gut dynamic balance.
