大鼠阶段性高脂饮食对后续个体肥胖及脂肪酸合成酶的影响 被引量:1

摘要 目的探讨大鼠阶段性高脂饮食对后续个体肥胖及相关蛋白脂肪酸合成酶(FAS)mRNA表达的影响,动态观察高脂饮食作用的持续性。方法Wistar雌性大鼠40只,受孕后随机分为高脂饮食组20只及正常饮食组20只,分娩后胎仔由妊娠期间正常饮食的母鼠喂哺,断乳后给予正常饮食至出生后8周,每组再分为两个亚组,分别给予高脂饮食和正常饮食,持续6周,记录每组的肥胖发生情况。分别在刚出生、出生后8周、出生后14周等不同时期处死动物,取样采用RT-PCR方法检测脂肪酸合成酶(FAS)m RNA的表达量。结果高脂组胎鼠在不同时期体质量均高于正常饮食组(P<0.05),高脂组胎鼠在不同时期检测FAS基因表达明显增强,与正常饮食组对比有统计学意义(P<0.05)。结论生命早期高脂饮食对后续个体肥胖相关基因表达有持续性影响,诱导肥胖的发生。 Objective To investigate the effect of high-fat diet on obesity and the expression of FAS in subsequent individuals,and to observe the persistence of the effect of high-fat diet.Methods Forty Wistar female rats were randomly divided into high fat diet group(20 rats)and normal diet group(20 rats).The offspring after delivery are fed by the normal diet of the mother mice during pregnancy,after weaning,normal diet was given until 8 weeks after birth,each group was further divided into two subgroups,each group was given a high-fat diet and a normal diet for 6 weeks,and the incidence of obesity in each group was recorded.The animals were executed at different stages of birth,8 weeks after birth and 14 weeks after birth,the expression of fatty acid synthase(FAS)was detected by RT-PCR.Results The weight of fetal rats in high fat group was higher than that in normal diet group at different periods(P<0.05).The expression of FAS gene in fetal mice of high fat group was significantly increased at different stages,which had statistical significance compared with the normal diet group(P<0.05).Conclusion High-fat diet in early life has a persistent effect on obesity-related gene expression in subsequent individuals and induces obesity.
