摘要
目的:研究二肽基肽酶4(DPP-4)抑制剂利格列汀对SD大鼠牙髓血运重建术后基质细胞衍生因子1(SDF-1)的表达及根管内新生组织的影响。方法:将90只大鼠随机分为实验组、对照组及空白组,每组30只。实验组灌胃利格列汀并对下颌第一磨牙进行血运重建术;对照组灌胃羧甲基纤维素钠(CMC-Na)并行血运重建术;空白组仅灌胃CMC-Na。术后1 d、4 d、7 d、14 d、21 d分别检测三组大鼠血清DPP-4活性及SDF-1浓度,行组织学观察新生组织形态及免疫组化染色观察根管内新生组织SDF-1的表达。结果:实验组术后5个时间点血清DPP-4活性均低于对照组及空白组(均P<0.05),而血清SDF-1浓度仅在术后1 d、4 d、7 d时高于对照组及空白组(均P<0.05)。组织学观察发现实验组、对照组术后1 d、4d、7 d时根管内大量炎症细胞浸润及红色基质样组织分布,14 d、21 d时炎症细胞减少,基质样组织红染变浅,形成空泡样结构。免疫组化结果显示三组术后1 d、4 d时根管内SDF-1表达量基本相同(P>0.05),实验组和对照组术后7 d、14 d,21 d时SDF-1表达均低于空白组(P<0.05)。结论:应用利格列汀能够抑制血清DPP-4活性,提高血清SDF-1浓度,但无法提高根管内SDF-1浓度,且利格列汀对血运重建术后根管内的新生组织形态无明显改善。
Objective: To study the effects of dipeptidyl peptidase 4(DPP-4)inhibitor--ligulitine--on the expression of matrix cell-derived factor 1(SDF-1)and new tissue in root canals after pulp revascularization in SD rats.Methods: Ninety rats were randomly divided into experimental group,control group,and blank group,with 30 rats in each group.The experimental group received liglitine by intragastric administration and performed revascularization on the mandibular first molar;the control group received sodium carboxymethylcellulose sodium(CMC-Na)by intragastrically and performed revascularization;the blank group received gavage CMC-Na only.On the day 1,day 4,day 7,day 14 and day 21 after operation,the activity of DPP-4 and the concentration of SDF-1 in serum of rats in the three groups were measured,and the morphology of newborn tissue was observed,and the expression of SDF-1 in root canal were observed by immunohistochemical staining.Results: The activity of serum DPP-4 in the experimental group was lower than that in the control group and the blank group at 5 time points after operation(all P<0.05),but the concentration of serum SDF-1 in the experimental group was only higher than that in the control group and the blank group on the day 1,day 4 and day 7 after operation(all P<0.05).Histological observation showed that there were a large number of inflammatory cells infiltration and red matrix-like tissue distribution in the root canal of the experimental group and the control group on the day 1,day 4 and day7 after operation,and the inflammatory cells decreased and the matrix-like tissue became lighter and formed a vacuole-like structure on the day 14 and day 21.The results of immunohistochemistry showed that the expression of SDF-1 in the root canals of the three groups were almost the same on the day 1 and day 4 after operation(P>0.05),while the expression of SDF-1 in the experimental group and the control group were lower than that of the blank group on the day 7 and day 21 after operation(P<0.05).Conclusion: Liglitine can inhibit the activity of serum DPP-4 and increase the concentration of serum SDF-1,but cannot increase the concentration of SDF-1 in the root canal,and there is no significant improvement in the morphology of new tissue in the root canal after revascularization.
